目的 探讨Ghrelin对内皮型一氧化氮合成酶(eNOS)活性的影响和作用机制.方法 分离培养大鼠主动脉血管内皮细胞并分为对照组(C组)和Ghrelin组.Ghrelin组加入终浓度为10、50、100和200nmol/L Ghrelin孵育3h收集标本.采用Western blot法测定内皮细胞eNOS和PKC蛋白表达水平.结果 与对照组相比,Ghrelin组eNOS磷酸化水平呈剂量依赖性增加(P<0.05),PKC磷酸化水平呈剂量依赖性降低(P<0.05).结论 Ghrelin可提高内皮细胞eNOS的活性,其机制可能与PKC通路的失活有关.

목적 탐토Ghrelin대내피형일양화담합성매(eNOS)활성적영향화작용궤제.방법 분리배양대서주동맥혈관내피세포병분위대조조(C조)화Ghrelin조.Ghrelin조가입종농도위10、50、100화200nmol/L Ghrelin부육3h수집표본.채용Western blot법측정내피세포eNOS화PKC단백표체수평.결과 여대조조상비,Ghrelin조eNOS린산화수평정제량의뢰성증가(P<0.05),PKC린산화수평정제량의뢰성강저(P<0.05).결론 Ghrelin가제고내피세포eNOS적활성,기궤제가능여PKC통로적실활유관.
Objective To investigate the effect of Ghrelin on endothelial nitric oxide synthase (eNOS) activity and its mechanism.Methods Vascular endothelial cells (ECs) were isolated from aorta in rats and were cultured.ECs were divided into two groups:control group and Ghrelin group.The cells were incubated with Ghrelin in different concentration (10,50,100 and 200 nmol/L) for 3 hour in Ghrelin group.The protein expression levels of nitric oxide synthase (eNOS) and protien kinase C (PKC) were measured by Western blot.Results Ghrelin can upregulate the phosphorylated eNOS expression level,and downregulate signal conduction pathway PKC phosphorylation in a dose dependent manner (P < 0.05).Conclusions Ghrelin can improve the activity of eNOS in ECs,and it may be related to the loss of PKC activity.
