目的观察凋亡相关斑点样蛋白(apoptosis-associated speck-like protein containing a CARD,ASC)和caspase-1在小鼠耳部皮肤急性光损伤模型中的表达并探讨其在急性光损伤中可能的作用机制.方法将10只小鼠分为5组,每组2只,分别对耳部照射30、90、150、300、600 mJ/cm2剂量的中波紫外线(ultraviolet B,UVB),测最小红斑量(minimal erythema dose,MED).将20只BALB/c小鼠随机分为正常对照组和照射4倍MED剂量UVB实验组,比较两组耳部皮肤临床和病理改变,免疫组化方法检测耳部皮肤ASC和caspase-1的表达.结果与对照组相比,实验组小鼠耳部皮肤出现红斑、水肿、毛细血管扩张.组织病理改变见表皮细胞水肿、核固缩,真皮血管扩张、管周淋巴细胞浸润等.免疫组化染色显示与对照组相比,实验组耳部表皮ASC和caspase-1表达增强,两组差异有统计学意义(t=65.0、56.5,P<0.01).结论 ASC和caspase-1在UVB致小鼠急性光损伤中具有一定的促损伤作用.

Objective To observe the expressions of apoptosis-associated speck (ASC)-like protein and caspase-1 in BALB/C mice following ultraviolet B (UVB)-induced acute light injury of skin and its possible mechanism. Methods Ten BALB/C mice were divided into five groups (2 in each group). Their ears were exposed to UVB irradiation at the doses of 30, 90, 150, 300, 600 mJ/cm2, respectively. The minimal erythema dose (MED) was measured. Twenty BALB/C mice were randomly divided into blank control group and experimental group (irradiated by 4 MED of UVB). Clinical and pathological changes of ear skin were compared between the two groups. Expressions of ASC-like protein and caspase-1 in ear skin were detected with immunohistochemical staining. Results Erythema, edema, telangiectasia, karyopyknosis, and perivascular lymphocytes infiltration occurred in experimental group but not in blank control group. Immunohistochemical staining showed that the expression levels of ASC-like protein and caspase-1 were significantly higher in experimental group than in blank control group (t=65.0 and 56.5, P<0.01). Conclusion ASC-like protein and caspase-1 are expressed in BALB/C mice following UVB-induced acute light injury.
