耐辐射奇球菌(D. radiodurans, DR)能在极端胁迫条件下生存,并且拥有一个独特的极端环境抗性基因组而被广泛用于研究.其中 DR1172是在耐辐射球菌中分离出的一段基因,与 LEA76家族同源,参与植物的抗旱机制.本研究利用基因工程手段以载体 pBI121为基础构建植物 DR1172-GV3103表达载体,然后利用花序浸染法将目的基因 DR1172转入模式植物拟南芥中,T2代得到50株植物,阳性为15棵,转化率为30%.该研究为探讨 DR1172基因的功能以及植物抗逆机制提供了丰富的拟南芥材料,而且对利用基因工程技术手段改良植物性状,培育出抗逆性强的植物优良品种有重要的指导价值.

DR1172 is a part of gene from D. radiodurans which has high resistance to radiation resistance. DR1172 and LEA76 are actually homologous in some way that they both participate in the drought resistant in plants. In this research, DR1172-GV3103 vector was constructed successfully and the DR1172 gene was inserted into Ara-bidopsis by using the transgenic technique. Transgenic Arabidopsis were identified by the molecular identification PCR. 15 positive plants were obtained from 50 T2 Arabidopsis and its conversion rate was 30%. The researches not only provide abundant plant resources to study the function of DR1172, but also have great important applica-tion value in plant gene engineering technology and cultivate quality variety of plants and agricultural production.
