通过单因素实验及正交试验对茶树菇多糖提取复合酶制剂进行筛选及工艺确定.复合酶制剂的筛选:通过单因素实验确定酶浓度,水解时间,初始pH及水解温度以及料液比对茶树菇水解液多糖含量的影响,并以此作为复合酶复配的依据.接着考察复合酶的浓度,水解初始pH,水解时间,水解温度和料液比对水解的影响,确定复合酶水解茶树菇提取多糖的最佳工艺.复合酶最优复配组合为纤维素酶∶木聚糖酶∶β-葡聚糖酶=3∶3∶6(酶活以80 U/mg计),最佳提取工艺为pH为5.0,酶解时间2 h,酶解温度50℃,料液比为1∶20,在此条件下多糖含量为197.65 mg/g,与其他方法相比有明显提高.

@@@@Screening of composite enzyme in extracting polysaccharides from Agrocybe aegirit and the optimal process were studied by both single-factor- and orthogonal experiments. Composite enzyme screening: the influence of enzyme concentration, pH, time, temperature and material/liquid ratio for extraction of polysaccharides from A. aegirit was studied by single-factor-experiments and component of composite enzyme was determined, then we investigated the composite enzyme concentration, pH, time, temperature and material liquid ratio for extraction of polysaccharides, determined the optimal process of extraction of polysaccharides from A.aegirit. The optimum conditions for extracting polysaccharides from A. aegirit were determined as follows:enzymatic treatment with Cellulase∶Xylanase∶β-Glucanase=3∶3∶6 using a material liquid ratio of 1∶20 at pH 5.0, 50℃for 2 hours resulted in the extraction of A. aegirit polysaccharides at a level of 197.65 mg/g;extraction of polysaccharide contents from A. aegirit by enzymatic digestion procedure resulted in better yields than other methods.
