建立一种快速测定百香果中绿原酸含量的方法.百香果果肉用微波辅助提取后,采用高效液相色谱法(HPLC)测定.色谱条件为:Eclipse XDB-C18色谱柱(4.6 mm×150 mm,5μm);流动相:A相为水+1%乙酸,B相为乙腈,采用梯度洗脱,最佳流速0.8 mL/min,检测波长320 nm,柱温40℃,进样量10μL.结果表明,绿原酸线性范围为19.80 mg/L~99.60 mg/L(R2=0.9997),平均加标回收率为97.68%(n=5),绿原酸平均含量为20.53 mg/g.该方法快速、准确,溶剂用量少,为快速测定百香果中的绿原酸提供参考.

@@@@A rapid method for the determination of chlorogenic acid in Passiflora eduis using high performance liquid chromatography (HPLC) method was established. The samples were extracted with method by microwave-assisted extraction and detected directly by HPLC method. The separation of chlorogenic acid was performed on Eclipse XDB-C18 column (4.6 mm×150 mm, 5μm), using acetonitrile-water as mobile phase with gradient elution, attheflowrateof0.8mL/min. The detection was carried out at 40℃with wavelength of 320 nm and injection volume of 10μL. Under the optimized conditions, the calibration curve of chlorogenic acid was linearly over the ranges of 19.80 mg/L-99.60 mg/L (R2=0.999 7). The average content of chlorogenic acid in Passiflora eduis samples was found to be 20.53 mg/g and an average recovery rate of 97.68%(n=5) was obtained. The proposed method has manyadvantagessuchashighsensitivity,fastness,aswellassmallamountsoforganicsolventconsuming.
