利用RP-HPLC测定小粒种咖啡不同部位(咖啡豆种仁、果皮和茎叶)中咖啡因含量.色谱柱:Agilent Eclips XDB-C18(Sun,4.6 mm×150 mm);流动相:甲醇-水(30∶70,体积比);流速:1 mL/min;检测波长:280 nm;柱温为30℃;咖啡因标准曲线方程为:Y=11.5662X+2.9827,R2=1.咖啡因在16μg/mL~80μg/mL范围内具有良好的线性关系.测定出攀枝花本地引种的小粒种咖啡豆中咖啡因含量为15.56 g/kg,果皮中含量为13.62 g/kg,茎叶中的含量为21.19 g/kg.精密度实验结果为咖啡豆种仁、果皮和茎叶中含量的RSD分别为1.01%、1.11%、1.59%;重复性实验结果为咖啡豆种仁、果皮和茎叶中咖啡因含量的RSD分别为0.69%、1.79%、1.38%;均小于2%,达到含量测定要求.咖啡豆种仁、果皮和茎叶中咖啡因回收率分别在95.6%~108.4%,94%~103.4%,96.5%~102.6%.
