目的观察丙酮酸乙酯对自身免疫性关节炎(CIA)大鼠关节炎症、骨质破坏及血清肿瘤坏死因子α(TNF-α)、白细胞介素10(IL-10)表达水平的影响,探讨其治疗类风湿关节炎的可能机制.方法实验大鼠分为正常对照组、CIA模型组和丙酮酸乙酯(EP)治疗组.用关节炎指数评价关节炎肿胀程度、HE染色、检测关节滑膜炎症及骨质破坏、酶联免疫吸附测定法(ELISA)检测血清TNF-α、IL-10浓度.结果 EP治疗组关节炎指数明显低于CIA模型组(P<0.05),EP治疗组炎性细胞渗出及骨质破坏明显低于CIA模型组(P<0.05),EP治疗组血清TNF-α水平较CIA模型组明显降低而IL-10水平明显升高(P<0.05).结论丙酮酸乙酯能减轻大鼠关节炎症、骨质破坏,能降低血清TNF-α表达水平、提高IL-10表达水平,表明丙酮酸乙酯有望成为治疗类风湿关节炎(RA)的一种新药.
目的觀察丙酮痠乙酯對自身免疫性關節炎(CIA)大鼠關節炎癥、骨質破壞及血清腫瘤壞死因子α(TNF-α)、白細胞介素10(IL-10)錶達水平的影響,探討其治療類風濕關節炎的可能機製.方法實驗大鼠分為正常對照組、CIA模型組和丙酮痠乙酯(EP)治療組.用關節炎指數評價關節炎腫脹程度、HE染色、檢測關節滑膜炎癥及骨質破壞、酶聯免疫吸附測定法(ELISA)檢測血清TNF-α、IL-10濃度.結果 EP治療組關節炎指數明顯低于CIA模型組(P<0.05),EP治療組炎性細胞滲齣及骨質破壞明顯低于CIA模型組(P<0.05),EP治療組血清TNF-α水平較CIA模型組明顯降低而IL-10水平明顯升高(P<0.05).結論丙酮痠乙酯能減輕大鼠關節炎癥、骨質破壞,能降低血清TNF-α錶達水平、提高IL-10錶達水平,錶明丙酮痠乙酯有望成為治療類風濕關節炎(RA)的一種新藥.
목적관찰병동산을지대자신면역성관절염(CIA)대서관절염증、골질파배급혈청종류배사인자α(TNF-α)、백세포개소10(IL-10)표체수평적영향,탐토기치료류풍습관절염적가능궤제.방법실험대서분위정상대조조、CIA모형조화병동산을지(EP)치료조.용관절염지수평개관절염종창정도、HE염색、검측관절활막염증급골질파배、매련면역흡부측정법(ELISA)검측혈청TNF-α、IL-10농도.결과 EP치료조관절염지수명현저우CIA모형조(P<0.05),EP치료조염성세포삼출급골질파배명현저우CIA모형조(P<0.05),EP치료조혈청TNF-α수평교CIA모형조명현강저이IL-10수평명현승고(P<0.05).결론병동산을지능감경대서관절염증、골질파배,능강저혈청TNF-α표체수평、제고IL-10표체수평,표명병동산을지유망성위치료류풍습관절염(RA)적일충신약.
Objective To observe the effect of ethyl pyruvate(EP)on synovial inflammation,bone degradation and serum TNF-α,IL-10 in rats with collagen-induced arthritis(CIA),and to explore its potential in treatment of rheumatoid arthritis(RA). Methods The rats were divided into normal control group,CIA model group and EP treatment group. The arthritis index were scored to evaluate arthritis swelling degree. The synovial inflammation and bone degradation in rats were determined by hematoxylin and eosin(HE)staining. The levels of TNF-αand IL-10 in serum were detected by enzyme-linked immunosorbent assay(ELISA). Result The EP treatment group had a lower index,synovium inflammation and bone destroy as compared with CIA group. The serum TNF-αlevel in the EP group was significantly lowered than those in CIA group,but the IL-10 level was remarkablely increased. Conclusion The ethyl pyruvate relieves joint swelling,synovial inflammation and bone degradation,decreases levels of TNF-α,IL-10 in CIA rats. Therefore,it may be a new drug for treatment of RA
