本研究采用免疫荧光及免疫组化DAB显色技术来检测Myogenin蛋白在五指山猪和长白猪胎儿和成年猪的骨骼肌中的定位和表达情况,并通过平均光密度统计分析Myogenin蛋白在五指山猪和长白猪胎儿和成年猪骨骼肌中表达水平.结果表明:Myogenin蛋白表达定位于骨骼肌细胞核表达;在五指山猪和长白猪胎儿和成年猪的腿部骨骼肌和背部骨骼肌中均检测到阳性产物,在猪胎儿期间骨骼肌中Myogenin蛋白的表达水平高于其在成熟猪骨骼肌中的表达;且在同种猪腿部骨骼肌中的表达水平显著高于其在背部骨骼肌中的表达;此外还检测到Myogenin蛋白在五指山猪骨骼肌中表达均显著高于其在长白猪骨骼肌中的表达(p<0.05).本研究得到的Myogenin蛋白在五指山猪和长白猪骨骼肌中存在显著的表达差异结果,为今后研究不同猪种间骨骼肌发育差异及探讨五指山猪矮小机制和发育机理提供了理论基础.
本研究採用免疫熒光及免疫組化DAB顯色技術來檢測Myogenin蛋白在五指山豬和長白豬胎兒和成年豬的骨骼肌中的定位和錶達情況,併通過平均光密度統計分析Myogenin蛋白在五指山豬和長白豬胎兒和成年豬骨骼肌中錶達水平.結果錶明:Myogenin蛋白錶達定位于骨骼肌細胞覈錶達;在五指山豬和長白豬胎兒和成年豬的腿部骨骼肌和揹部骨骼肌中均檢測到暘性產物,在豬胎兒期間骨骼肌中Myogenin蛋白的錶達水平高于其在成熟豬骨骼肌中的錶達;且在同種豬腿部骨骼肌中的錶達水平顯著高于其在揹部骨骼肌中的錶達;此外還檢測到Myogenin蛋白在五指山豬骨骼肌中錶達均顯著高于其在長白豬骨骼肌中的錶達(p<0.05).本研究得到的Myogenin蛋白在五指山豬和長白豬骨骼肌中存在顯著的錶達差異結果,為今後研究不同豬種間骨骼肌髮育差異及探討五指山豬矮小機製和髮育機理提供瞭理論基礎.

The immunofluorescence and immunohistochemistry DAB color method were used to detect the expression of Myogenin protein of Wuzhishan pig and Landrace pig's skeletal muscle in fetal period and mature stage, and the IOD of the statistical analysis used to analyze the Myogenin protein expression level of Wuzhishan pig and Landrace pig's skeletal muscle in fetal period and mature stage. The results showed that Myogenin protein was expressed in nucleus of skeletal muscle. Myogenin protein expressed in skeletal muscle in fetal period and mature stage of Wuzhishan pig and Landrace pig. The higher expression level of Myogenin protein in skeletal muscle in fetal period than it in mature stage, the higher expression level of Myogenin protein in leg skeletal muscle than it in back skeletal muscle, and in addition detected that the higher expression level of Myogenin protein in Wuzhishan pig than it in landrace pig (p<0.05). Myogenin protein expression was significantly different in skeletal muscle of Wuzhishan pig and landrace pig. The results provided a theoretical basis for future study on the dwarf mechanism and development of Wuzhishan pig.
