目的建立提取分离川芎有效成分的方法,研究提取物的凝血作用.方法以阿魏酸含量为指标,用HPLC测定川芎中阿魏酸的含量,采用正交试验优化不同提取分离的参数等.结果渗漉优于热回流方法;渗漉提取的最佳方案为4倍50%乙醇,渗漉速率1 mL/kg;川芎提取物高剂量组0.32 g/kg连续灌胃给药10 d,可显著延长小鼠的凝血时间.结论该提取工艺设计合理,结果可靠,为开发川芎抗凝血活性部位提供了实验参考.

@@@@Objective To establish a method for the extraction of active components of Chuanxiong Rhizoma, and investigate the blood coagulation effect of the Chuanxiong Rhizoma extract. Methods Conditions for the extraction were studied by orthogonal experimental design, and the contents of ferulic acid were measured by HPLC. Results Percolation method was better than thermal reflux method. The best conditions of percolation extraction were as follows: 4-fold amount of 50% alcohol, the percolation rate of 1mL/kg. High-dose (0.32g/kg) of Chuanxiong Rhizoma extract significantly prolonged the clotting time in bleeding mice for continuous intragastric administration of 10 days. Conclusion This extraction technology is feasible and applicable, which will provide the experimental reference for the development of anticoagulated blood active site of Chuanxiong Rhizoma.
