以莲藕的顶芽和侧芽为外植体,接种于不同外源激素配比的MS培养基上,诱导不定芽,并进行分化培养.结果显示:不定芽诱导的最佳培养基为:MS +1.0 mg /L 6-BA +0.15 mg /L NAA +0.2%Gelrite,其诱导率为93.3%,不定芽平均数为2.14;不定芽分化的最佳培养基为MS +1.0 mg /L 6-BA +0.05 mg/L NAA,其存活率为83.3%,芽长平均值为0.98 cm.其中,6-BA在莲藕不定芽诱导及分化中起重要作用,但选择适当浓度是得到高质量芽的关键.以Gelrite代替琼脂粉作为固化剂,可明显改善外植体的褐化现象,若添加活性炭则效果更佳.

@@@@With the apical shoots and lateral buds of lotus root as explants , MS mediums with different concentration of plant hormones were used to induce generation and differentiation of adventitious buds .The results showed that the optimum formula of medium for adventitious buds inducement was MS +1.0 mg/L6 -BA +0.15mg/LNAA+0.2%Gelrite, the ratio of inducement was 93.3%and the average number of buds was 2.14.The most ideal medium for adventitious buds differentiation was MS +1.0 mg/L6 -BA +0.05 mg/L NAA, the ratio of surviv-al was 83.3%and the average length of buds was 0.98 cm.6-BA played an important role in the induction and differentiation of lotus root, but the choice of appropriate concentration was the key to get high quality buds .Gel-rite, instead of Agar, added in medium as curing agent can obviously improve the browning of explants .If activated carbon was added, the effect would be better.
