目的通过建立脾细胞移植和脾片联合移植动物模型来观察脾片移植早期细胞免疫功能恢复状况.方法 Wistar大鼠48只,随机分为4组,A组为对照组,行脾切除术,B组为脾片移植组,C组为脾细胞移植组,D组为脾细胞脾片联合移植组,在建立模型前及模型成功后1周、4周、8周抽血,检测外周血T细胞亚群CD3+、CD4+、CD8+及CD4+/CD8+水平并进行比较.结果 A组脾切除后1、4、8周CD3+、CD4+、CD8+降低(P<0.05),B组移植后1、4周CD3+、CD4+、CD8+与术前相比降低(P<0.05),至8周逐渐接近正常(P>0.05),C组移植后1周CD3+、CD4+、CD8+与术前相比降低(P<0.05),至4周逐渐接近正常(P>0.05),而8周逐渐降低(P<0.05),D组术后1周CD3+、CD4+、CD8+与术前相比降低(P<0.05),4周和8周接近正常(P>0.05);CD3+、CD4+、CD8+在术后1、4周C和D组与A和B组有差异(P<0.05),在术后8周B和D组与A和C组有差异(P<0.05).结论在脾片移植的基础上联合脾细胞移植能弥补脾片移植早期的部分细胞免疫功能.

목적통과건립비세포이식화비편연합이식동물모형래관찰비편이식조기세포면역공능회복상황.방법 Wistar대서48지,수궤분위4조,A조위대조조,행비절제술,B조위비편이식조,C조위비세포이식조,D조위비세포비편연합이식조,재건립모형전급모형성공후1주、4주、8주추혈,검측외주혈T세포아군CD3+、CD4+、CD8+급CD4+/CD8+수평병진행비교.결과 A조비절제후1、4、8주CD3+、CD4+、CD8+강저(P<0.05),B조이식후1、4주CD3+、CD4+、CD8+여술전상비강저(P<0.05),지8주축점접근정상(P>0.05),C조이식후1주CD3+、CD4+、CD8+여술전상비강저(P<0.05),지4주축점접근정상(P>0.05),이8주축점강저(P<0.05),D조술후1주CD3+、CD4+、CD8+여술전상비강저(P<0.05),4주화8주접근정상(P>0.05);CD3+、CD4+、CD8+재술후1、4주C화D조여A화B조유차이(P<0.05),재술후8주B화D조여A화C조유차이(P<0.05).결론재비편이식적기출상연합비세포이식능미보비편이식조기적부분세포면역공능.
Objective To observe functional restoration of pristine cellular immunity after splenic sliices autotransplantation by establishing animal model on combination trantran-splantation of antogeneic Splenic cell and slices. Methods 48 wistar rats are randomly divid ed into four groups, A group: splenectomy. B group: splenic slices transplantation. C group: spleen cell transplantation. D group: combination transplantation. Before animal moddel and within 1,4,8 weeks after stablishing animal model, peripheral blood is drewn and detected the level of peripheral blood T-cyte subset CD3+,CD4+,CD8+ and CD4+/CD8+, At the same time, the results are compared. Result In A group postoperative CD3+,CD4+,CD8+ became lower(P<0.05); In B group CD3+,CD4+,CD8+ became lower compared with preoperative levels at the postoperative 1th and 4th week(P<0.05), got close common at the postoperative 8th week(P>0.05); In C group CD3+,CD4+,CD8+ became lower compared with preoperative levels at the postoperative 1th week(P<0.05), got close common at the postoperative 4th week(P>0.05), but became gradually lower at the postoperative 8th week(P<0.05); In D group CD3+,CD4+,CD8+ became lower compared with preoperative levels at the postoperative 1th week(P<0.05), got close common at the postoperative 4th and 8th week(P>0.05); Statistically difference was detected between the C, D and A, B groups for CD3+,CD4+,CD8+ at the postoperative 1th and 4th week(P<0.05), Statistically difference was detected between the B, D and A, C groups for CD3+,CD4+,CD8+ at the postoperative 8th week(P<0.05). Conclusion Combination trantransplantation of antogeneic Splenic cell and slices can retrieve partial pristinel cellular immune function.
