2BFI%Experimental autoimmune encephalomyelitisrats (EAE)%Matrix metal oproteinase-9 (MMP-9)
目的观察咪唑啉2受体配体(2BFI)对实验性自身免疫性脑脊髓炎(EAE)小鼠的行为、病理和脑脊髓中基质金属蛋白酶-9(MMP-9)表达的影响.方法用髓鞘少突胶质细胞糖蛋白(MOG)免疫C57BL/6小鼠制作EAE模型,将小鼠随机分为5组:药物干预组(EAE+2BFI 5 mg·kg-1组、EAE+2BFI 10 mg·kg-1组、EAE+2BFI 20 mg·kg-1组)、阳性对照组(EAE+NS组)、正常对照组(完全福氏佐剂+NS组),观察比较各组行为学和病理组织学的变化,通过RT-PCR技术测定大脑和脊髓MMP-9 mRNA水平,并用免疫组化方法观测2BFI对大脑组织MMP-9阳性表达的影响.结果与EAE+NS组比,2BFI 10、20 mg·kg-1药物干预组的最高行为学评分和病理变化均有减轻;而且三种不同剂量2BFI干预组大脑和脊髓组织MMP-9mRNA水平和大脑组织中MMP-9阳性表达均较EAE+NS组明显降低.结论2BFI对EAE有保护作用,可能与其调节抑制MMP-9,保护血脑屏障,减少炎症细胞通过血脑屏障有关.

@@@@Objective To investigate the effect of 2BFI on the behavior and MMP-9 expression in experimental autoim-mune encephalomyelitis(EAE) mice, Methods C57B/L mice were immunized with myelin oligodendrocyte glycoprotein (MOG) to establish the mice model of EAE. The mice were randomly divided into 5 groups:EAE+2BFI groups (2BFI 5 mg·kg-1 , 10 mg·kg-1 and 20 mg·kg-1), EAE+NS group and CFA+NS group. After immunization, the mice in EAE+2BFI groups were treated with in-traperitoneal injection of 2BFI 5 mg·kg-1 , 10 mg·kg-1 and 20 mg·kg-1, respectively;whereas the mice in EAE+NS and CFA+NS groups were treated with an intraperitoneal injection of 0.1ml normal saline. The immunohistochemical staining was used to detect the expression of MMP-9 protein and RT-PCR was used to determine MMP-9 mRNA expression. Results In 2BFI 10 and 20 mg·kg-1 groups, the maximum clinical scores and pathological change were both decreased. Compare with EAE+NS Group, the expression of MMP-9 protein and mRNA in the brain of EAE mice was decreased in three EAE+2BFI groups. Conclusion 2BFI can protect mice from EAE, which is associated with reduced MMP-9 expression in central nervous system.
