目的:探讨知母总皂苷(total saponins from Rhizoma Anemarrhenae,TS)对链脲霉素(streptozotocin, STZ)诱导的糖尿病(diabetes mellitus,DM)大鼠海马中乙酰胆碱酯酶(acetylcholinesterase,AChE)活性的影响及可能机制.方法:雄性 Sprague Dawley 大鼠单次腹腔注射(ip)STZ(55 mg·kg-1)诱导 DM 大鼠模型后,按血糖值随机分为 DM 组和 TS 组,TS 组灌胃(ig)给予 TS(200 mg·kg-1),每天一次,连续8周.测定海马中 AChE活性、丙二醛(malondialdehyde,MDA)和还原型谷胱甘肽(reduced glutathione,GSH)水平,以及血清中 MDA、GSH 和葡萄糖水平.结果:相对于正常大鼠,8周病程的 DM 大鼠海马中 AChE 活性显著增加,海马和血清中MDA 水平显著增加、GSH 明显降低,给予 TS 干预后能够改善 AChE 的变化,显著降低 MDA 水平,增加 GSH水平.此外,TS 还能够明显降低 DM 大鼠的空腹血糖和增加体重.结论:知母总皂苷对糖尿病状态下海马中AChE 活性具有一定的改善作用,可能与其抗氧化应激和改善 DM 基本症状有关.

목적:탐토지모총조감(total saponins from Rhizoma Anemarrhenae,TS)대련뇨매소(streptozotocin, STZ)유도적당뇨병(diabetes mellitus,DM)대서해마중을선담감지매(acetylcholinesterase,AChE)활성적영향급가능궤제.방법:웅성 Sprague Dawley 대서단차복강주사(ip)STZ(55 mg·kg-1)유도 DM 대서모형후,안혈당치수궤분위 DM 조화 TS 조,TS 조관위(ig)급여 TS(200 mg·kg-1),매천일차,련속8주.측정해마중 AChE활성、병이철(malondialdehyde,MDA)화환원형곡광감태(reduced glutathione,GSH)수평,이급혈청중 MDA、GSH 화포도당수평.결과:상대우정상대서,8주병정적 DM 대서해마중 AChE 활성현저증가,해마화혈청중MDA 수평현저증가、GSH 명현강저,급여 TS 간예후능구개선 AChE 적변화,현저강저 MDA 수평,증가 GSH수평.차외,TS 환능구명현강저 DM 대서적공복혈당화증가체중.결론:지모총조감대당뇨병상태하해마중AChE 활성구유일정적개선작용,가능여기항양화응격화개선 DM 기본증상유관.
Objective:To investigate the effects of total saponins from Rhizoma Anemarrhenae(TS)on acetylcholinesterase(AChE)activity in the hippocampus of streptozotocin-induced diabetic rats and their possible mechanisms. Methods:Male Sprague Dawley rate were subjected to a single intraperitoneal injection of streptozotocin(STZ),55mg·kg-1 introduced diabetic rats,which were randomly divided into two groups according to blood glucose values. The TS group rats were treated with TS(200 mg·kg-1,ig)for 8 weeks,the diabetic control rates received normal saline solution and their blood glucose and body weight were measured monthly and weekly,respectively. The hippocampus was obtained for assays of AChE activity as well as levels of malondialdehyde(MDA)and reduced glutathione(GSH);blood was collected for assays of levels of MDA, GSH and glucose. Results:Compared with that of the age-matched normal rats, AChE activity was markedly increased in the hippocampus of diabetic rats,and TS treatment blocked such an increase. Meanwhile, diabetes significantly elevated the MDA level and reduced GSH level both in the hippocampus and the serum,and TS treatment significantly prevented such changes. In addition,TS significantly decreased the fasting blood glucose level and increased the body weights of diabetic rats. Conclusion:TS has an inhibitory effect on AChE activity in the hippocampus in diabetic rats,which is at least partly due to its antioxidant and anti-diabetic activity.
