目的:探讨缺氧缺血性脑损伤大鼠脑组织中SDF-1与NSCs各时间点表达情况和作用.方法:72只7日龄SD大鼠随机分为对照组36只和模型组36只.模型组参照Rice法单侧结扎新生大鼠左颈总动脉复制HIBD动物模型,对照组只切开颈部正中皮肤,分离但不结扎左侧颈总动脉.处死前2小时腹腔注射BrdU,对照组和模型组分别在3小时,1天,3天,7天,10天,14天6个时间点处死6只,采用HE染色观察脑组织变化情况,免疫组织化学染色观察海马区SDF-1和BrdU的表达情况.结果:HIBD组SDF-1在1天时表达开始明显升高,3天达到高峰,到7天时略有点降低,组内比较差异性显著,有统计学意义(p<0.05),组间各时间点差异显著,有统计学意义(p<0.05),对照组各时间点差异无显著,无统计学意义(p>0.05).BrdU表达在1天表达很少,在3天表达增高,7天表达开始下降,14天表达更少.IODBrdU阳性细胞/IODSDF-1阳性细胞的均值比在3天左右最大.结论:SDF-1可能趋化内源性神经干细胞修复脑损伤,并且在3天左右时能够更大程度修复脑损伤.

Objective: To examine the expression of stromal cell-derived factor-1(SDF-1)and endogenous neural stem cells (NSCs) in cerebral tissue of neonatal rats with hypoxic-ischemic brain damage in each time point and to discuss the relationship between them. Methods: 72 seven-age’s SD rats were randomly divided into the control group with only 36 and the model group witn the rest. A rat model of hypoxic-ischemic brain damage was established by clipping left cephalic artery referring to Rice method;the control group were only cut the center skin of cervical part open, segregated but not clipped left cephalic artery. The control group and the model group respectively in 3h, 1d, 3d, 7d, 10d, 14d six time point only 6 will be put to death two hours before BrdU intraperitoneal injection, HE used to observe changes in brain tissue staining and immunohistochemical staining to observe the hippocampus SDF-1 and BrdU expression. Results: HIBD group SDF-1 expression in 1d began to rise significantly, 3d peak, to 7d slightly a little lower. There was significant difference within the group and was statistical significance (p<0.05).Among Each time point groups, there was statistically significant(p<0.05).In each time point the control group there was no significant difference, no statistical significance (p>0.05). BrdU expression in 1d was very few, in 3d heighten, 7d began to decline, 14d less. The ratio of the IOD of positive cell of BrdU and SDF-1 expression was the maximum on the third day about. Conclusion SDF-1 may attract endogenous neural stem cells to repair brain injury, and in the 3 d around to a greater degree.
