目的体外培养大鼠胎盘间充质干细胞,大鼠脊髓匀浆上清液诱导胎盘间充质干细胞向神经细胞分化.方法消化大鼠胎盘组织、贴壁培养获得间充质干细胞,大鼠脊髓匀浆上清液诱导胎盘间充质干细胞分化,采用免疫细胞化学的方法对分化的细胞进行鉴定.结果胎盘组织中分离出的间充质干细胞与骨髓间充质干细胞有相似形态和细胞表面标志,并在脊髓匀浆的诱导下可诱导表达神经元和星型胶质细胞标志:神经元特异性烯醇化酶(NSE)、神经微丝(NF)和胶质纤维酸性蛋白(GFAP).结论胎盘间充质干细胞能分化为神经元样细胞,脊髓匀浆可以作为一种有效的诱导剂将胎盘间充质干细胞诱导分化为神经细胞.
目的體外培養大鼠胎盤間充質榦細胞,大鼠脊髓勻漿上清液誘導胎盤間充質榦細胞嚮神經細胞分化.方法消化大鼠胎盤組織、貼壁培養穫得間充質榦細胞,大鼠脊髓勻漿上清液誘導胎盤間充質榦細胞分化,採用免疫細胞化學的方法對分化的細胞進行鑒定.結果胎盤組織中分離齣的間充質榦細胞與骨髓間充質榦細胞有相似形態和細胞錶麵標誌,併在脊髓勻漿的誘導下可誘導錶達神經元和星型膠質細胞標誌:神經元特異性烯醇化酶(NSE)、神經微絲(NF)和膠質纖維痠性蛋白(GFAP).結論胎盤間充質榦細胞能分化為神經元樣細胞,脊髓勻漿可以作為一種有效的誘導劑將胎盤間充質榦細胞誘導分化為神經細胞.
목적체외배양대서태반간충질간세포,대서척수균장상청액유도태반간충질간세포향신경세포분화.방법소화대서태반조직、첩벽배양획득간충질간세포,대서척수균장상청액유도태반간충질간세포분화,채용면역세포화학적방법대분화적세포진행감정.결과태반조직중분리출적간충질간세포여골수간충질간세포유상사형태화세포표면표지,병재척수균장적유도하가유도표체신경원화성형효질세포표지:신경원특이성희순화매(NSE)、신경미사(NF)화효질섬유산성단백(GFAP).결론태반간충질간세포능분화위신경원양세포,척수균장가이작위일충유효적유도제장태반간충질간세포유도분화위신경세포.
@@@@Objective To culture placental-derived mesenchymal stem cells from rat in vitro,and to observe differentiation of the placenta-derived mesenchymal stem cells into neural cells induced by the rat 's spinal cord homoge-nate.Methods Rat's placental tissue was digested and adherent cultured to obtain mesenchymal stem cells ,placental-derived mesenchymal stem cells were induced by the rat 's spinal cord homogenate,and immunocytochemical methods was used for identification of differentiated cells .Results Placental-derived mesenchymal stem cells and bone mesenchymal stem cells have similar morphology and cell surface markers ,and induced by the spinal cord homogenate ,can express markers of neurons and astrocytes:neuron-specific enolase sign(NSE),neurofilament(NF) and glial fibrillary acidic pro-tein(GFAP).Conclusion Placental mesenchymal stem cells can differentiate into neuron -like cells,and spinal cord ho-mogenate can induce the placenta-derived mesenchymal stem cells to differentiate into neural cells .
