目的观察红花注射液(ICT)对2,4,6-三硝基苯磺酸(2,4,6-TNBS)所致大鼠溃疡性结肠炎(UC)的疗效及其对结肠组织中 EGF 和 TGF-β1表达的影响.方法将大鼠随机分为3组:模型组、ICT 组和正常对照组(n 均为10).用 TNBS 灌肠复制大鼠 UC 模型,用 ICT 干预治疗,观察大鼠疾病活动指数(DAI)及大肠组织学损伤以判断疗效;用免疫组化法检测 UC 大鼠结肠黏膜 EGF 和 TGF-β1的表达,用逆转录-聚合酶链反应(RT-PCR)法检测 EGF mRNA 和 TGF-β1mRNA 的表达,从而探讨其治疗机制.结果与模型组相比,ICT 组的DAI 及组织学损伤显著改善(P <0.05);与正常对照组相比,ICT 组与模型组大鼠结肠组织中 EGF,TGF-β1蛋白及其 mRNA 的表达均显著增高(P <0.05);而与模型组相比,ICT 组结肠组织中 EGF 及其 mRNA 的表达均显著增高(P <0.05);TGF-β1蛋白及其 mRNA 的表达显著降低(P <0.05).结论 ICT 对 TNBS 所致大鼠 UC 具有一定疗效,ICT 可以增加 EGF,TGF-β1的表达,促进创伤组织的修复,是 ICT 治疗溃疡性结肠炎可能的作用机制之一.

목적관찰홍화주사액(ICT)대2,4,6-삼초기분광산(2,4,6-TNBS)소치대서궤양성결장염(UC)적료효급기대결장조직중 EGF 화 TGF-β1표체적영향.방법장대서수궤분위3조:모형조、ICT 조화정상대조조(n 균위10).용 TNBS 관장복제대서 UC 모형,용 ICT 간예치료,관찰대서질병활동지수(DAI)급대장조직학손상이판단료효;용면역조화법검측 UC 대서결장점막 EGF 화 TGF-β1적표체,용역전록-취합매련반응(RT-PCR)법검측 EGF mRNA 화 TGF-β1mRNA 적표체,종이탐토기치료궤제.결과여모형조상비,ICT 조적DAI 급조직학손상현저개선(P <0.05);여정상대조조상비,ICT 조여모형조대서결장조직중 EGF,TGF-β1단백급기 mRNA 적표체균현저증고(P <0.05);이여모형조상비,ICT 조결장조직중 EGF 급기 mRNA 적표체균현저증고(P <0.05);TGF-β1단백급기 mRNA 적표체현저강저(P <0.05).결론 ICT 대 TNBS 소치대서 UC 구유일정료효,ICT 가이증가 EGF,TGF-β1적표체,촉진창상조직적수복,시 ICT 치료궤양성결장염가능적작용궤제지일.
@@@@Objective To investigate the therapeutic effects of injection of carthamus tinctorius (ICT) on ulcerative colitis(UC) induced by 2,4,6-trinitro -ben-zene-sulfonic acid(TNBS) in rats as well as its effect on the expression of epidermal growth factor (EGF) and trans forming growth factor-β1(TGF-β1).Methods Thirty male Wistar rats were randomly divided into treatment group ,TNBS control group and normal control group(n =10).The rats in treatment group were treated with injection of carthamus tinctorius (ICT).Curative effect was judged by investigate the disease activity index (DAI) and histological damage.The expression of EGF and TGF-β1 in colon tissue was detec-ted by immunohistochemistry(IHC).And the expression of EGFmRNA and TGF -β1mRNA were detected by reverse transcription -polymerase chain reaction(RT-PCR) method.Results Compared with model group,the DAI and histological damage of colon tissues in treatment group were significantly improved.The level of EGF and TGF-β1 in model group and treatment group were significantly higher than that in normal control group(P <0.05).The level of EGF and TGF-β1 in treatment group was significantly higher than that in model group (P <0.05).The level of TGF-β1 in treatment group was lower than that in model group (P <0.05).Conclusion This study indicates that the ICT can relieve colitis in rats induced by TNBS and increasing EGF and decreasing TGF -β1 may be one of its functional mechanism .
