目的研究男性不育症患者精液细胞成分DNA的病理改变.方法采用流式细胞术和常规精液检查法,对146例不育男性患者和30例正常生育男性进行精液细胞DNA分析.结果正常生育男性精液细胞成分主要为单倍体成熟精子(91.72±2.60)%.男性不育症患者精液细胞成分等显示多种变化:(1)精液中主要为单倍体成熟精子.(2)单倍体的成熟精子数量减少.(3)单倍体圆形精子细胞增多.(4)单倍体精子峰增宽.(5)≥2倍体的细胞(白细胞、G 1期精原细胞、精母细胞、上皮细胞及4倍体细胞等)增多.(6)亚单倍体细胞碎片和凋亡细胞增多.结论精液细胞DNA变化反映男性不育症患者精液细胞成分有明显改变.精液DNA流式细胞术分析可作为研究和诊断男性不育症的辅助方法.

Objective To study the pathological changes of semen DNA of infertile males. Methods Semen sample from 146 patients with infertility and 30 fertile men were detected by conventional assays and semen DAN was analyzed by flow cytometry (FCM). The relative proportion and pathological changes of various cells in the semen were investigated. Results The number of mature haploid spermatozoa(91.72±2.60)%in semem of normal male were observed. Various pathological changes in semen of the patients with male infertility were found including the most amount of mature haploid spermatozoa in semen ,decrease in the number of mature haploid spermatozoa, round haploid spermatids increasing, the peak of haplois sematozoa widned, the number of diploid cells(Leukocytes, G1-spermatogomia, etc) and tetraploid cells increasing, Sub-haploid debris and apoptosis cells increasing. Conclusion Obvious pathological changes in semen cells were defined in the patients with infertility. Semen DNA analysis can be an auxiliary method for the study and diagnosis of male infertility.
