目的探讨酒精对大鼠心肌组织自噬相关基因及蛋白表达的影响.方法雄性SD大鼠20只,随机分为饮酒组和对照组,用酒精灌胃的方式建立饮酒模型,并用RT-PCR方法检测两组心肌组织中Beclin-1和LC3ⅡmRNA的表达,用Western Blot方法检测相应蛋白水平的表达,用透射电镜进行形态学观察,并检测血清中心肌损伤标志物的变化.结果饮酒组与对照组比较自噬基因Beclin-1及LC3Ⅱ的表达增加(P<0.01),相应蛋白表达也升高,电镜下可见自噬小体生成,血清中心肌损伤标志物明显升高.结论饮酒可引起大鼠心肌组织自噬相关基因Beclin-1和LC3Ⅱ及蛋白表达增加.

Objective To investigate the effects of drinking alcohol on the expression of autophagy related genes in rat myocardium.Methods Twenty male SD rats were randomly divided into 2 groups:control group(n=10) and alcohol feeding(AF) group(n=10).The mRNA and protein expression of autophagy related genes were determined by RT-PCR and Western Blot,the autophagosomes were observed via Transmission Electron Microscopy,myocardial damage markers were assayed via Automatic biochemical analyzer.Results The expression of mRNA and protein of Beclin-1(P<0.01) and LC3Ⅱ(P<0.01) was increased significantly in the AF group compared with the control group.The typical autophagosome structure was observed via the Transmission Electron Microscopy in AF group.Myocardial damage markers were obviously raised in AF group.Conclusion Alcohol upregulates the mRNA and protein expression of autophagy related genes in myocardia of rats.
