目的探讨内皮细胞蛋白C受体(EPCR)在人脐静脉内皮细胞(ECV304)上的表达及炎症因子白介素-1β(IL-1β),PPARγ激动剂曲格列酮对其影响.方法采用流式细胞仪检测ECV304中EPCR的表达;实时定量PCR技术(Real-Time PCR)检测IL-1β作用后ECV304细胞EPCR mRNA表达;曲格列酮和IL-1β共同作用于ECV304细胞,实时定量PCR技术检测EPCR mRNA表达.结果 EPCR在ECV304中有很高表达,IL-1β下调ECV304细胞EPCR的表达,而曲格列酮可以明显改善IL-1β对EPCR的抑制.结论 ECV304中EPCR的表达可受到炎症因子IL-1β的影响,曲格列酮对上述影响有一定的拮抗作用,对ECV304损伤有保护作用.

Objective To investigate the effect of IL-1β on Endothelial cell protein C receptor(EPCR) expression in umbilical vein endothelial cells.Methods EPCR expression was assessed by flow cytometry in umbilical vein endothelial cells.The effects of IL-1β and troiglitazone were measured by Real-Time PCR. Results Umbilical vein endothelial cells have a high expression of EPCR.IL-1β reduce EPCR expression.And troglitazone could significantly improve the inhibition.Conclusion We observed EPCR expression in umbilical vein endothelial cells in vitro.Some factors such as IL-1β can impact on EPCR.However,troglitazone had protective effects of EPCR on injured cells.
