目的:探讨血清视黄醇结合蛋白4(RBP4)诱导人脐静脉内皮细胞(HUVEC)炎症的作用机制.方法:采用MTT还原法检测细胞活性,通过DCFH-DA检测活性氧的生成,Western blotting分析相关蛋白的表达,凝胶滞留法(EMSA)分析NF-kB的DNA结合活性.结果:RBP4处理后,MTT检测发现人脐静脉内皮细胞存活率显著低于对照组,流式细胞仪的结果表明活性氧的水平也显著升高.结论:RBP4可诱导人脐静脉内皮细胞(HUVEC)毒性及炎症反应.

목적:탐토혈청시황순결합단백4(RBP4)유도인제정맥내피세포(HUVEC)염증적작용궤제.방법:채용MTT환원법검측세포활성,통과DCFH-DA검측활성양적생성,Western blotting분석상관단백적표체,응효체류법(EMSA)분석NF-kB적DNA결합활성.결과:RBP4처리후,MTT검측발현인제정맥내피세포존활솔현저저우대조조,류식세포의적결과표명활성양적수평야현저승고.결론:RBP4가유도인제정맥내피세포(HUVEC)독성급염증반응.
@@@@Objective:To investigate the mechanism of action of serum retinol binding protein 4 (RBP4) in human umbilical vein endothelial cells (HUVEC) induced inflammation. Methods:MTT reduction assay cell activity by DCFH-DA to detect reactive oxygen species generation, Western blotting analysis of related proteins, gel retardation (EMSA) analysis of NF-kB DNA binding activity. Results:RBP4 in treatment, MTT assay found that human umbilical vein endothelial cells was significantly lower than the control group, the flow cytometry results show that reactive oxygen species levels were also significantly elevated. Conclusion:RBP4 in human umbilical vein endothelial cells (HUVEC) could induce toxicity and inflammatory response.
