目的:观察龙葵氯仿提取物对人肺腺癌A549细胞凋亡的影响.方法:设龙葵氯仿提取物低、中、高三个剂量组(分别以1mg/mL浓度50、300、500μL加药)和空白对照组,运用流式细胞术观察其结果.结果:与空白对照组比较,不同剂量的龙葵氯仿提取物作用于A549细胞24h后,早期凋亡细胞百分比依次增至4.6%、16.8%和23.6%;晚期凋亡细胞及坏死细胞百分比依次增至1.1%、1.1%和28.9%.其中龙葵提取物高剂量组,早期凋亡细胞比例和晚期凋亡细胞及坏死细胞比例均明显高于空白对照组(P<0.01).结论:龙葵氯仿提取物可诱导A549细胞凋亡并呈剂量依赖性;诱导肿瘤细胞凋亡和坏死可能是龙葵氯仿提取物抑制肿瘤细胞增殖的主要作用机制之一.

Objective: To observe the influence of chloroform extract from Solanum nigrum L on human lung ade?nocarcinoma A549 cell apoptosis. Methods: Low, middle, and high dosage groups of chloroform extract from Sola?num nigrum L(50, 300, and 500μg) and control groups were set and flow cytometry was used to observe cell apoptosis. Results: Compared with the control group, after being treated with different dosages of chloroform ex?tract from Solanum nigrum L, the percentage of early apoptotic A549 cells increased to 4.6%,16.8%,and 23.6%, and the percentage of late apoptotic cells /necrotic cells increased to 1.1%, 1.1%, and 28.9%. A significant dif?ference in the percentages of cell apoptosis was noted between the high dosage group of chloroform extract from Solanum nigrum L and the control group (P<0.01). Conclusion: Chloroform extract from Solanum nigrum L can induce apoptosis of A549 cells, and showed a dose-effect relationship. Induction of tumor cell apoptosis and ne?crosis may be one of the main mechanisms of the extract on inhibiting proliferation of tumor cells.
