目的:观察乌司他丁对局灶性脑缺血再灌注损伤大鼠的肿瘤坏死因子(TNF-α)表达的变化,探讨其对缺血性脑损伤的保护机制.方法:采用线栓法制备大鼠大脑中动脉栓塞的局灶性脑缺血再灌注模型,实验动物随机分为假手术组、脑缺血再灌注模型组、药物治疗组.应用免疫组织化学法和半定量 RT-PCR 法检测各组缺血皮质中 TNF-α表达的动态变化.结果:乌司他丁组缺血皮质中 TNF-α的表达与模型组相比明显下调且差异有统计学意义.结论:乌司他丁通过抑制 TNF-α的表达可改善脑缺血大鼠的神经功能.
目的:觀察烏司他丁對跼竈性腦缺血再灌註損傷大鼠的腫瘤壞死因子(TNF-α)錶達的變化,探討其對缺血性腦損傷的保護機製.方法:採用線栓法製備大鼠大腦中動脈栓塞的跼竈性腦缺血再灌註模型,實驗動物隨機分為假手術組、腦缺血再灌註模型組、藥物治療組.應用免疫組織化學法和半定量 RT-PCR 法檢測各組缺血皮質中 TNF-α錶達的動態變化.結果:烏司他丁組缺血皮質中 TNF-α的錶達與模型組相比明顯下調且差異有統計學意義.結論:烏司他丁通過抑製 TNF-α的錶達可改善腦缺血大鼠的神經功能.
목적:관찰오사타정대국조성뇌결혈재관주손상대서적종류배사인자(TNF-α)표체적변화,탐토기대결혈성뇌손상적보호궤제.방법:채용선전법제비대서대뇌중동맥전새적국조성뇌결혈재관주모형,실험동물수궤분위가수술조、뇌결혈재관주모형조、약물치료조.응용면역조직화학법화반정량 RT-PCR 법검측각조결혈피질중 TNF-α표체적동태변화.결과:오사타정조결혈피질중 TNF-α적표체여모형조상비명현하조차차이유통계학의의.결론:오사타정통과억제 TNF-α적표체가개선뇌결혈대서적신경공능.
Objective:To observe the effect of ulinastatin on the expression of TNF-αon focal cerebral ischemia-reperfusion injury in rats, so that probe the mechanism of ulinastatin against cerebral ischemia injury. Methods:Ischemia was induced by temporary middle cerebral artery occlusion. Male adult Sprague-Dawley rats were randomly divided into the sham- operated group, middle cerebral artery occlusion reperfusion injury model group,and ulinastatin -treated group. Rats were killed to detect the expression of TNF-α by immunohistochemistry and RT-PCR. Results:The expression of TNF-αin cortex of ulinastatin -treated group was lower than that in the model group .Conclusions:These data suggest that ulinastatin can improve neurological function after cerebral ischemia -reperfusion by decreasing the expression of TNF-αin the injured region.
