以小鼠为动物模型,通过直接灌胃的方法,验证不同剂量林蛙油抗氧化、抗衰老的作用;利用Real time PCR的方法分析小鼠肝脏组织中与抗氧化相关基因Cu/ZnSOD的时空表达规律.结果表明,5周龄组和15月龄组小鼠灌胃林蛙油后,肝组织中Cu/ZnSOD基因表达量显著增加,但总体上看,5周龄鼠0.45 g·kg-1剂量组Cu/ZnSOD基因表达增加量要普遍高于2.25 g·kg-1剂量组,但与1.35 g·kg-1剂量组差异不显著;而15月龄鼠1.35 g·kg-1剂量组Cu/ZnSOD基因表达增加量要显著高于0.45和2.25 g·kg-1剂量组,但0.45和2.25 g·kg-1剂量组差异不显著;而且15月龄组Cu/ZnSOD基因表达量升高的速度较慢,一般在灌胃第30天或第45天才出现显著差异.

In the current study, the anti-oxidant and anti-aging effects of Rana chensinensis oil were verified by using mice as animal models and feeding them with the oil directly. The expression profiling of the mouse liver antioxidant-related Cu/ZnSOD gene was monitored by Realtime PCR. The results showed that by directly feeding with Rana chensinensis oil, the expression of Cu/ZnSOD gene in mouse liver tissue was significantly highly up-regulated in both 5 weeks and 15 months mice.Generally, the Cu/ZnSOD gene expression was higher in 15 month mice with the dosages of 0.45 g·kg-1 than the dosage of 2.25 g·kg-1, but it was not significantly with the dosages of 1.35 g·kg-1. The Cu/ZnSOD gene expression was higher in old mice with the dosages of 1.35 g·kg-1 than the dosage of 0.45 and 2.25 g· kg-1, but the dosage of 0.45 g·kg-1 was not significantly with the dosages of 2.25 g·kg-1. The up-regulation of Cu/ZnSOD gene expression was slower in 15 month mice than 5 weeks ones. The significant difference in 15 month mice was only able to be observed after 30 or 45 d feeding treatments.
