利用方波脉冲电场增强乳酸菌细胞通透性,通过将乳酸菌细胞离心,制成浓度为108 cfu·mL-1菌悬液,采用不同方波电脉冲条件对其进行处理.结果表明,在0.5 mol·L-1蔗糖的等渗脉冲介质中,电场强度2.25 kV·cm-1,脉冲宽度15μs,脉冲数5,间隔时间2 s,此时,乳酸菌细胞的存活率平均为78.24%,细胞内平均荧光强度为5480.表明脉冲电场可有效增强乳酸菌细胞通透性.
利用方波脈遲電場增彊乳痠菌細胞通透性,通過將乳痠菌細胞離心,製成濃度為108 cfu·mL-1菌懸液,採用不同方波電脈遲條件對其進行處理.結果錶明,在0.5 mol·L-1蔗糖的等滲脈遲介質中,電場彊度2.25 kV·cm-1,脈遲寬度15μs,脈遲數5,間隔時間2 s,此時,乳痠菌細胞的存活率平均為78.24%,細胞內平均熒光彊度為5480.錶明脈遲電場可有效增彊乳痠菌細胞通透性.
이용방파맥충전장증강유산균세포통투성,통과장유산균세포리심,제성농도위108 cfu·mL-1균현액,채용불동방파전맥충조건대기진행처리.결과표명,재0.5 mol·L-1자당적등삼맥충개질중,전장강도2.25 kV·cm-1,맥충관도15μs,맥충수5,간격시간2 s,차시,유산균세포적존활솔평균위78.24%,세포내평균형광강도위5480.표명맥충전장가유효증강유산균세포통투성.
In order to increase the permeability of Lactobacillus cells by square-wave pulses. Lactobacillus cells was centrifugated and suspended in the medium with a rather high concentration about 108 cfu·mL-1. Suspension of lactobacillus cells was exposed to square-wave electric pulses in different conditions. Results showed that fraction survival cells was 78.24% and fluorescence intensity was 5 480 on average, when we treated the lactobacillus cells under the optimal condition that was electric field strength 2.25 kV·cm-1, pulse durations 15μs, pulse numbers 5, and time interval 2 s, in the isotonic buffer solution with concentration of sucrose (0.5 mol·L-1). This study indicated that permeability of Lactobacillus cells can be increased by square-wave pulses.
