目的研究猪苓对脂多糖(LPS)诱导的J 774细胞白细胞介素6(IL-6)与iNOS表达的影响.方法体外培养J 774细胞,用不同浓度的猪苓进行干预,MTT法测定猪苓对J 774细胞活力的影响;利用LPS(10μg/mL)刺激细胞建立炎症模型,经不同浓度猪苓进行干预后,RT-PCR法检测细胞中IL-6、iNOS mRNA的表达.结果猪苓在0.05~50μg/mL剂量范围内对J 774细胞无明显细胞毒性作用.猪苓可明显抑制LPS诱导的J 774细胞IL-6、iNOS mRNA的表达,并具有一定的量效关系.结论猪苓呈浓度依赖性抑制LPS诱导的J 774细胞IL-6、iNOS的表达,从而发挥抗炎作用.

목적연구저령대지다당(LPS)유도적J 774세포백세포개소6(IL-6)여iNOS표체적영향.방법체외배양J 774세포,용불동농도적저령진행간예,MTT법측정저령대J 774세포활력적영향;이용LPS(10μg/mL)자격세포건립염증모형,경불동농도저령진행간예후,RT-PCR법검측세포중IL-6、iNOS mRNA적표체.결과저령재0.05~50μg/mL제량범위내대J 774세포무명현세포독성작용.저령가명현억제LPS유도적J 774세포IL-6、iNOS mRNA적표체,병구유일정적량효관계.결론저령정농도의뢰성억제LPS유도적J 774세포IL-6、iNOS적표체,종이발휘항염작용.
Objective To observe the effects of polyporus umbellatus on the expression of interleukin 6 and iNOS from J 774 cells induced by LPS. Methods J 774 cells were cultivated and treated with polyporus umbellatus. The effect of polyporus umbellatus on cell vitality of J 774 cells was measured by MTT assay. J 774 cells were stimulated by LPS and treated with polyporus umbellatus. Real-time quantitative PCR(RT-PCR) was used to detect the expression level of IL-6,iNOS mRNA. Results The cell vitality of Polyporus umbellatus group had no obvious different compared with control group in the 0.5-50 μg/mL dose range;polyporus umbellatus could inhibit the content of IL-6 and iNOS mRNA. Conclusion Polyporus umbellatus could inhibit IL-6 and iNOS release induced by LPS in a dose-dependent manner.
