目的评价七氟烷预处理联合后处理对大鼠心肌缺血再灌注时血红素氧合酶-1(HO-1)表达的影响.方法雄性SD大鼠40只,体重250~280 g,随机分为5组(n=8):假手术组(S组)、心肌缺血再灌注组(I/R组)、七氟烷预处理组(S-Pre组)、七氟烷后处理组(S-Post组)、七氟烷预处理联合后处理组(S-Pre+S-Post组).采用结扎左冠状动脉前降支30 min,再灌注120 min的方法制备大鼠心肌缺血再灌注模型.于再灌注120 min时,检测血清CK-MB、心肌梗死面和心肌HO-1的表达.结果与S组比较,其余4组血清CK-MB、心肌梗死面积和心肌组织HO-1表达均增加(P<0.05);与I/R组比较,S-Pre组、S-Post组和S-Pre+S-Post组血清CK-MB和心肌梗死面积降低,心肌组织HO-1表达增高(P<0.05);与S-Pre组和S-Post组比较,S-Pre+S-Post组血清CK-MB和心肌梗死面积降低,心肌组织HO-1表达增高(P<0.05).结论与单纯七氟烷预处理或后处理比较,两种方法联合应用可增加HO-1的表达,从而进一步减轻了大鼠心肌缺血再灌注损伤.

목적평개칠불완예처리연합후처리대대서심기결혈재관주시혈홍소양합매-1(HO-1)표체적영향.방법웅성SD대서40지,체중250~280 g,수궤분위5조(n=8):가수술조(S조)、심기결혈재관주조(I/R조)、칠불완예처리조(S-Pre조)、칠불완후처리조(S-Post조)、칠불완예처리연합후처리조(S-Pre+S-Post조).채용결찰좌관상동맥전강지30 min,재관주120 min적방법제비대서심기결혈재관주모형.우재관주120 min시,검측혈청CK-MB、심기경사면화심기HO-1적표체.결과여S조비교,기여4조혈청CK-MB、심기경사면적화심기조직HO-1표체균증가(P<0.05);여I/R조비교,S-Pre조、S-Post조화S-Pre+S-Post조혈청CK-MB화심기경사면적강저,심기조직HO-1표체증고(P<0.05);여S-Pre조화S-Post조비교,S-Pre+S-Post조혈청CK-MB화심기경사면적강저,심기조직HO-1표체증고(P<0.05).결론여단순칠불완예처리혹후처리비교,량충방법연합응용가증가HO-1적표체,종이진일보감경료대서심기결혈재관주손상.
Objective To investigate the effects of sevoflurane preconditioning combined with postconditioning on HO-1 during myocardial ischemia-reperfusion(I/R)in rat. Methods Forty male SD rats weighing 250~280g were randomLy divided into 5 groups (n=8 each):Sham operation group (group S), Ischemia and reperfusion group (group I/R), Sevoflurane preconditioning group (group S-Pre), Sevoflurane postconditioning group (group S-Post),Sevoflurane preconditioning combined with postconditioning group (group S-Pre+S-Post).Established the model of rats′myocardial ischemia reperfusion,ligation the left anterior descending coronary artery in 30 min, 120 min after reperfusion.After being reperfused for 120 minutes,determination of blood serum CK-MB level and myocardial infarction area,myocardial HO-1 expression were detected. Results Compared with the group S,after reperfusion 120 min,the levels of CK-MB,myocardial infarction area and HO-1 expression were increased in the rest of the four groups (P<0.05);Compared with the I/R group, the levels of CK-MB and myocardial infarction area were decreased(P<0.05)in group S-Pre, group S-Post and group S-Pre+S-Post. While HO-1 expression were increased (P<0.05).Compared with the group S-Pre and group S-Post,the levels of CK-MB and myocardial infarction area were decreased(P<0.05)in group S-Pre+S-Post.While HO-1 expression were increased (P<0.05).Conclusion Compared with only sevoflurane preconditioning or postconditioning, the two methods in combination can reduce HO-1 expression,and further reduce myocardial ischemia–reperfusion injury.
