目的探讨丙泊酚对肝癌细胞株SMMC-7721侵袭特性和MMP-2、MMP-9的影响.方法不同浓度的丙泊酚处理SMMC-7721细胞,用Transwell小室模型测定SMMC-7721对细胞外基质Matrigel的侵袭力,明胶酶谱法检测培养上清液中MMP-2、MMP-9的变化.结果经过5μg/mL、10μg/mL、20μg/mL的丙泊酚处理后,SMMC-7721侵袭力均明显下调,培养上清液中MMP-2、MMP-9的含量也明显下降.结论丙泊酚可抑制肝癌细胞株SMMC-7721的体外侵袭力,同时明显降低SMMC-7721细胞分泌基质金属蛋白酶.

[ABSTRACT] Objective To study the anti-invasion effect of propofol on liver cancer cell and expression of MMP-2 and MMP-9. Methods Propofol-treated SMMC-7721 invasion to matrigel were detected by transwell chamber method. The contents of MMP-2 and MMP-9 in culture supernatant were measured by gelatin zymography. Results Propofol inhibited SMMC-7721 invasion to the matrigel at the concentration of 5μg/mL, 10μg/mL and 20μg/mL. Meanwhile, the concentrations of MMP-2 and MMP-9 were significantly decreased in matrix. Conclusion Propofol could significantly decrease the invasion effect and metalloproteinase expression of SMMC-7721.
