以韭菜“汉中冬韭”品种为试材,本试验筛选了种子消毒的最佳方法和不定芽生根的最佳培养基,研究了不同激素组合对韭菜根尖不定芽再生的影响.结果表明,韭菜种子表面消毒采用70%乙醇浸泡30 s,再用0.1%升汞消毒10 min为宜;以根尖为外植体,诱芽的最佳BA、NAA培养基组合为MS+BA 2 mg/L+NAA 1 mg/L,诱芽率为45.33%;如果加入KT,最佳组合为MS+BA 1 mg/L+NAA 1 mg/L+KT 1 mg/L,诱愈率为50%,芽分化率为38.89%.不定芽生根的最佳培养基为MS+IAA 0.3 mg/L,IAA浓度过大,反而抑制生根.另外,不同基因型的芽分化频率略有差别,以“汉中冬韭”品种最好.

Using Chinese chive“Hanzhong Dongjiu”as the material, the experiments were carried out to study the effects of different hormone combination on the regeneration of adventitious buds from the root tips and to screen the best seeds disinfection method, as well as the best medium formulas for rooting of adventitious buds. the results showed that the suitable sterilization for the seed surface of chinese chive should be treated in 70%ethanol for 30 s, prior to treatment in 0.1%HgCl solution for 10 min;the best medium formula for inducing callus and adventitious buds from the root explants was MS+BA 2 mg/L+NAA 1 mg/L, which the buds induction rate rea-ched to 45.33%. If the KT was introduced, the best medium formula was MS+BA 1 mg/L+NAA 1 mg/L+KT 1 mg/L, which the ratio of the calli and buds induction were 50%and 38.89%respectively;The best medium fomula for rooting of adventitious buds was MS+IAA 0.3 mg/L. If the concentration of IAA was too high, the IAA would inhibits rooting of the bud. In addition, the ratios of buds induction for different genotype were a little different, the“Hanzhong Dingjiu”was the best among a few varieties.
