The recent discovery of several potent antitumor antibiotics containing enediynes or analogues and models of enediyne has pushed the exploration of enediynes to the forefront of chemical and biological research. Not only their unique structural features, But the most fascinating aspect is their unprecedented mechanism of the cleavage DNA that originates from the chemical reactivity of the enediyne moiety. Novel and more efficient approaches to the enediyne functionality were also discussed.
