Combretastatin A-4(CA-4)是从植物Combretum caffrum中分离出来的一种活性成分,具有抗肿瘤和抗血管生成作用,是一种很有潜力的抗癌药物.CA-4对很多癌细胞有良好的抑制效果,包括一些多药耐药性癌细胞,而且对增生扩散的血管内皮细胞有很好的选择性作用.本文对CA-4的构效关系和近年来的发展进行了阐述.

Combretastatin A-4(CA-4) is a potent antineoplastic and antiangiogenesis natural substance isolated from the South African tree Combretum caffrum. CA-4 shows strong cytotoxicity against a variety of cancer cells, including multi-drug resistant cancer cell lines. It has also been demonstrated to exert highly selective effects in proliferating endothelial cells. In this paper, the structure-activity relationships and pharmacological properties of the CA-4 derivatives as a class of potent anticancer agents are reviewed and discussed.
