以固定化人工膜为载体研究胰蛋白酶的动态固定化,考察缓冲液种类(硼酸盐、磷酸盐和Tris?HCl)、浓度、pH对胰蛋白酶固载率和酶活性的影响.结果显示,pH=8.0的10mmol/L Tris?HCl缓冲液是胰蛋白酶的优化固载条件.在此条件下,初步研究了动态固定化胰蛋白酶酶解器,发现其具有较高酶活性,且胰蛋白酶抑制剂能对其活性进行特异性抑制.

In this study, Immobilized Artificial Membrane (IAM) was chosen as matrix and trypsin was used as immobilized macromolecule. The effect of different buffer solutions (Borate, Phosphate, and Tris?HCl), concentrations, and pH on the immobilizing rate and activity of the immobilized trypsin was investigated. The optimal condition of off-line immobilization was Tris?HCl buffer (10 mmol/L, pH 8.0). On the basis of the obtained optimal condition, the trypsin enzyme reactor was prepared, and the preliminary result showed that the dynamically immobilized enzyme reactor possessed high activity, which can be specifically inhibited by the trypsin inhibitor.
