CT-1 gp130%Hypertension%Rats
目的观察 CT-1mRNA 和 gp130在高血压大鼠心肌中的表达变化、意义,以及替米沙坦对心肌营养素-1(CT-1)和糖蛋白130(gp130)表达的影响.方法20只雄性 SD 大鼠行腹主动脉缩窄术后2周造成压力负荷性心肌肥厚模型,随机分为心肌肥厚组和替米沙坦组,另设10只作为假手术组.替米沙坦组灌胃给药3 mg/(kg·d),连续4周.原位杂交法检测心肌的 CT-1mRNA 表达水平;免疫组化法检测心肌中 gp130表达水平.结果与假手术组比较,心肌肥厚组心肌组织 CT-1mRNA 和 gp130表达水平明显增加.与心肌肥厚组比较,替米沙坦组 CT-1mRNA 和 gp130含量显著降低(P<0.01).结论压力超负荷性大鼠心肌中CT-1mRNA 及其受体 gp130的过度表达与心室重构的发生密切相关;替米沙坦逆转心室重构的机制可能与抑制 CT-1及受体 gp130的过度表达相关.

목적관찰 CT-1mRNA 화 gp130재고혈압대서심기중적표체변화、의의,이급체미사탄대심기영양소-1(CT-1)화당단백130(gp130)표체적영향.방법20지웅성 SD 대서행복주동맥축착술후2주조성압력부하성심기비후모형,수궤분위심기비후조화체미사탄조,령설10지작위가수술조.체미사탄조관위급약3 mg/(kg·d),련속4주.원위잡교법검측심기적 CT-1mRNA 표체수평;면역조화법검측심기중 gp130표체수평.결과여가수술조비교,심기비후조심기조직 CT-1mRNA 화 gp130표체수평명현증가.여심기비후조비교,체미사탄조 CT-1mRNA 화 gp130함량현저강저(P<0.01).결론압력초부하성대서심기중CT-1mRNA 급기수체 gp130적과도표체여심실중구적발생밀절상관;체미사탄역전심실중구적궤제가능여억제 CT-1급수체 gp130적과도표체상관.
Objective To determine the changes of expression of CT-1mRNA and GP130 in myocardium and significance in hypertensive rats. Methods 20 male SD rats were randomly divided into 2 groups at two weeks after partial ligating aorta: hypertrophied group(LVH,n=10)and telmisartan group(Tel,n=10).Sham-operated group(Sham, n=10)was selected to serve non-partial ligating aorta. Telmisartan group were delivered by direct gastric gavage [(3 mg/(kg·d))for 4 weeks. AngⅡlevel in myocardium was measured by radioimmunoassay. The expression of CT-1mRNA of cardiomyocytes was determined with in site hybridization histochemistry. The protein level of gp130 of cardiomyocytes was determined by immunohistochemistry analysis..Results Compared with sham-operated group, Cardiotrophin-1mRNA gene expression and the protein expressing of gp130 in cardiac tissue were significant enhanced (P<0.01)in the LVH. Conclusion Telmisartan down-regulated the cardiotrophin-1 mRNA and the protein expression of gp130 in the elmisartan roup.
