目的以SD大鼠的脂肪干细胞(ADSCs)作为研究对象,在体外探讨研究基质细胞衍生因子(SDF-1)对大鼠源性的脂肪干细胞(ADSCs)增殖、粘附方面的影响.方法分别用不同浓度的SDF-1刺激AD-SCs,观察48h,用MTT法检测不同浓度的SDF-1对ADSCs增殖的影响以及利用黏附模型评价不同浓度SDF-1对ADSCs粘附能力影响.结果 SDF-1剂量依赖性增强ADSCs的增殖能力在100mg/L时达到顶峰,与对照组相比,不同浓度SDF-1处理的ADSCs其粘附能力呈现浓度依赖性.结论 SDF-1可以促进ADSCs的黏附,但是只有在一定剂量范围内的SDF-1才可以促进ADSCs的增殖.

Objective To investigate the effect of stromal cell-derived factor-1 on proliferation and adhesion of ADSCs using the rat adipose-derived stem cells(ADSCs) as the research objects.Methods Different concentration of SDF-1 were used to stimulate ADSCs respectively .After 48 hours,the proliferation of ADSCs was detected with MTT method,and the adhesion ability of ADSCs was analyzed by microscope .Results SDF-1 increased the proliferation of ADSCs in a dose-dependent manner,but its role of promoting effect reached the top at 100mg/L and down-regulation at 200mg/L.Conclution SDF-1 can improve the proliferation of ADSCs in a certain concentration range .SDF-1 increased the adhesion of ADSCs in a dose-dependent manner.
