背景:自体骨髓间充质干细胞体外诱导培养后的多方向分化潜能,体内植入能否修复股骨头坏死成为临床骨科的一个研究热点.目的:柚皮甙诱导的骨髓间充质干细胞修复兔股骨头坏死头的作用.方法:柚皮甙诱导兔骨髓间充质干细胞向成骨方向分化,用自制同种异体松质骨载体负载,制备兔股骨头坏死模型,造模后第3周将12只成年新西兰大白兔随机数字表法分为2组,实验组左侧置入空白载体,对照组左侧不植入任何物品;两组右侧均置入复合有诱导后骨髓间充质干细胞的载体.结果与结论:植入后8周植入物表面有大量骨痂形成,与骨端连接紧密,少部形成连续性骨桥.12周部分充填区广布相对较成熟的骨小梁,但较正常骨小梁粗、排列不够规则,可见缺损区完全被新骨代替,提示柚皮甙诱导的骨髓间充质干细胞可修复兔股骨头坏死.
揹景:自體骨髓間充質榦細胞體外誘導培養後的多方嚮分化潛能,體內植入能否脩複股骨頭壞死成為臨床骨科的一箇研究熱點.目的:柚皮甙誘導的骨髓間充質榦細胞脩複兔股骨頭壞死頭的作用.方法:柚皮甙誘導兔骨髓間充質榦細胞嚮成骨方嚮分化,用自製同種異體鬆質骨載體負載,製備兔股骨頭壞死模型,造模後第3週將12隻成年新西蘭大白兔隨機數字錶法分為2組,實驗組左側置入空白載體,對照組左側不植入任何物品;兩組右側均置入複閤有誘導後骨髓間充質榦細胞的載體.結果與結論:植入後8週植入物錶麵有大量骨痂形成,與骨耑連接緊密,少部形成連續性骨橋.12週部分充填區廣佈相對較成熟的骨小樑,但較正常骨小樑粗、排列不夠規則,可見缺損區完全被新骨代替,提示柚皮甙誘導的骨髓間充質榦細胞可脩複兔股骨頭壞死.

BACKGROUND:Autologous bone marrow mesenchymal stem cel s exhibit multipotential differentiation after in vitro culture. There is increasing concern that whether transplantation of autologous bone marrow mesenchymal stem cel s can be used for repair of femoral head necrosis. OBJECTIVE:To investigate the mechanism by which naringin induces bone marrow mesenchymal stem cel s to repair femoral head necrosis in rabbits. METHODS:Rabbit bone marrow mesenchymal stem cel s were induced with naringin to differentiate into osteoblasts and then loaded onto cancel ous bone al ograft. Femoral head necrosis rabbit models were prepared. At 3 weeks after femoral head necrosis induction, 12 adult New Zealand rabbits were randomly divided into two groups. In the experimental group, blank cancel ous bone al ograft was implanted into the left femoral head necrosis region. Identical y, nothing was implanted into the left femoral head necrosis region in the control group. The right femoral head necrosis region in each group was implanted with cancel ous bone al ograft loaded by naringin induced bone marrow mesenchymal stem cel s. RESULTS AND CONCLUTION:At 8 weeks after implantation, a large amount of cal us formed on the surface of the al ograft and connected tightly with bone ends, and continuous“bone bridge”formed in some parts. At 12 weeks, relatively mature bone trabeculae were observed in the fil ing area, which were thicker, and arranged more irregularly compared with normal bone trabeculae. Bone defect region was completely covered by the newly formed bone. These findings suggest that naringin-induced bone marrow mesenchymal stem cel s can be used for repair of femoral head necrosis in rabbits.
