背景:以自体骨髓间充质干细胞和软骨细胞作为种子细胞修复软骨缺损可达到理想效果,但存在细胞数量不足及二次创伤等问题.
目的:观察同种异体骨髓间充质干细胞和软骨细胞与聚乙酸-聚乙醇酸共聚物复合修复关节软骨的可行性.
方法:将15只新西兰大白兔随机分为实验组、对照组、空白组,制作关节软骨缺损模型,分别于骨缺损处植入同种异体骨髓间充质干细胞和同种异体软骨细胞与聚乙酸-聚乙醇酸共聚物复合体、自体骨髓间充质干细胞和自体软骨细胞与聚乙酸-聚乙醇酸共聚物复合体、聚乙酸-聚乙醇酸共聚物材料.
结果与结论:术后12周苏木精-伊红及Masson三色染色显示,实验组软骨缺损处可见软骨细胞,呈圆形或多角形,柱状排列,软骨陷窝形成明显,可见大量细胞外基质沉积,修复组织显示出透明软骨样,与周围软骨结合好,与底层骨结合紧密;对照组与实验组无明显差别;空白组软骨缺损处可见纤维样细胞.说明同种异体骨髓间充质干细胞和软骨细胞与聚乙酸-聚乙醇酸共聚物复合可修复关节软骨缺损.
揹景:以自體骨髓間充質榦細胞和軟骨細胞作為種子細胞脩複軟骨缺損可達到理想效果,但存在細胞數量不足及二次創傷等問題.

結果與結論:術後12週囌木精-伊紅及Masson三色染色顯示,實驗組軟骨缺損處可見軟骨細胞,呈圓形或多角形,柱狀排列,軟骨陷窩形成明顯,可見大量細胞外基質沉積,脩複組織顯示齣透明軟骨樣,與週圍軟骨結閤好,與底層骨結閤緊密;對照組與實驗組無明顯差彆;空白組軟骨缺損處可見纖維樣細胞.說明同種異體骨髓間充質榦細胞和軟骨細胞與聚乙痠-聚乙醇痠共聚物複閤可脩複關節軟骨缺損.

방법:장15지신서란대백토수궤분위실험조、대조조、공백조,제작관절연골결손모형,분별우골결손처식입동충이체골수간충질간세포화동충이체연골세포여취을산-취을순산공취물복합체、자체골수간충질간세포화자체연골세포여취을산-취을순산공취물복합체、취을산-취을순산공취물재료.
결과여결론:술후12주소목정-이홍급Masson삼색염색현시,실험조연골결손처가견연골세포,정원형혹다각형,주상배렬,연골함와형성명현,가견대량세포외기질침적,수복조직현시출투명연골양,여주위연골결합호,여저층골결합긴밀;대조조여실험조무명현차별;공백조연골결손처가견섬유양세포.설명동충이체골수간충질간세포화연골세포여취을산-취을순산공취물복합가수복관절연골결손.
BACKGROUND:The autologous bone marrow mesenchymal stem cel s and chondrocytes as seed cel s can achieve the ideal effect for repairing the articular cartilage defect, but the shortage of seed cel s and secondary trauma are difficult problems for researchers.
@@@@OBJECTIVE:To investigate the feasibility for repairing the articular cartilage defect by co-culture of al ogenic bone marrow mesenchymal stem cel s and al ogenic chondrocytes mixed with poly lactic-co-glycolic acid.
@@@@METHODS:Fifteen New Zealand rabbits were divided into three groups, including experimental group, control group and blank group, and the articular cartilage defect models were prepared. The articular cartilage defect was fil ed with al ogenic bone marrow mesenchymal stem cel s and the complex of al ogenic chondrocytes and poly lactic-co-glycolic acid, autologous bone marrow mesenchymal stem cel s and the complex of autologous chondrocytes and poly lactic-co-glycolic acid, and the poly lactic-co-glycolic acid copolymer material.
@@@@RESULTS AND CONCLUSION:Hematoxylin-eosin and Masson trichrome staining at 12 weeks after operation showed that the chondrocytes could be seen in the articular cartilage defect of the experimental group with round or polygonal-shaped and in a columnar arrangement. The typical cartilage lacunae were formed and a large number of extracel ular matrix depositions could be seen and the repaired tissue showed a hyaline cartilage-like and integrated wel with the surrounding tissues and closed wel with underlying bone. There was no significant difference between the experimental group and the control group;in the blank group, the fiber-like cel s could be seen in the articular cartilage defect. It is feasible to repair articular cartilage defect by co-culture of al ogenic bone marrow mesenchymal stem cel s and al ogenic chondrocytes mixed with poly lactic-co-glycolic acid.
