目的探讨TNFα、NF-κB和PPAR-γ等在MODS大鼠外周血中的相互关系.方法脂多糖(LPS)静脉注射法制造大鼠脓毒症模型,分别在第1天、5天、10天、12天用酶联免疫吸附剂测定法检测外周血中TNF-α的水平,外周血单个核细胞内转录因子PPARγ、NF-κB p65表达.结果 TNFα和NF-κB在模型组大鼠外周血中,第1天表达逐渐增强,第5天起高水平表达一直持续至12天(P<0.01).PPAR-γ在对照组和第1天模型组外周血中表达最强,第5天表达开始减弱.TNFα和NF-κB在外周血的表达呈明显正相关关系,与PPAR-γ则呈负相关关系(P<0.01).结论 TNFα和NF-κB与大鼠脓毒血症程度呈正相关,而 PPAR-γ则相反.

Objective To investigate the TNFα, NF-κB and PPAR-γ, such as MODS in rat peripheral blood of mutual relations. Methods lipopolysaccharide (LPS) intravenous injection of rat sepsis model of manufacturing, in the 1st day, 5 days, 10 days, 12 days by enzyme-linked immunosorbent assay detected in peripheral blood levels of TNF-α, Peripheral blood mononuclear cels with transcription factors PPARγ, NF-κB p65 expression. TNFα and the results of NF-κB in the rat model of peripheral blood, the 1st day of expression gradualy increased, the 5th day of a high level of expression until 12 days (P <0.01). PPAR-γ in the control group and 1 day model group of peripheral blood to express the strongest, the 5th day of expression began to decline. TNFα and NF-κB in the peripheral blood showed a positive correlation between clear, and PPAR-γ is a negative correlation (P<0.01). Conclusion TNFα and NF-κB and peripheral blood disease have a good degree of consistency in the relationship, PPAR-γ on the contrary.
