目的观察坐骨神经慢性压迫性损伤(chronic constriction injury of the sciatic nerve, CCI)对大鼠背根神经节(dorsal root ganglion, DRG)中 Fc 段γ受体Ⅰ型(Fc-gamma receptor type Ⅰ, FcγR Ⅰ)表达的影响,初步探讨其参与神经病理性疼痛的机制.方法35只雌性 Wistar 大鼠随机分为7组(n=5):空白对照组(Control)、假手术组(Sham 3d,7d,14d 组)、手术组(CCI3d,7d,14d 组).分别于术前、术后1d,3d,7d,10d,14d 测定各组大鼠热/机械痛阈,并用 Western blotting方法检测术后各规定时间点 DRG 中 FcγR Ⅰ表达变化.结果 Control 组和 Sham 组间热/机械痛阈和 FcγR Ⅰ蛋白表达无统计学差异(P >0.05),CCI 组大鼠热/机械痛阈明显下降(P <0.01)伴 DRG 中 FcγR Ⅰ表达量增多(P <0.05).结论 DRG 中 FcγR Ⅰ蛋白表达上调可能参与神经元兴奋性的提高,在神经病理性疼痛的形成与维持中发挥重要作用.

목적관찰좌골신경만성압박성손상(chronic constriction injury of the sciatic nerve, CCI)대대서배근신경절(dorsal root ganglion, DRG)중 Fc 단γ수체Ⅰ형(Fc-gamma receptor type Ⅰ, FcγR Ⅰ)표체적영향,초보탐토기삼여신경병이성동통적궤제.방법35지자성 Wistar 대서수궤분위7조(n=5):공백대조조(Control)、가수술조(Sham 3d,7d,14d 조)、수술조(CCI3d,7d,14d 조).분별우술전、술후1d,3d,7d,10d,14d 측정각조대서열/궤계통역,병용 Western blotting방법검측술후각규정시간점 DRG 중 FcγR Ⅰ표체변화.결과 Control 조화 Sham 조간열/궤계통역화 FcγR Ⅰ단백표체무통계학차이(P >0.05),CCI 조대서열/궤계통역명현하강(P <0.01)반 DRG 중 FcγR Ⅰ표체량증다(P <0.05).결론 DRG 중 FcγR Ⅰ단백표체상조가능삼여신경원흥강성적제고,재신경병이성동통적형성여유지중발휘중요작용.
Objective To detect the effects of chronic constriction injury of the sciatic nerve (CCI) on Fc-gamma receptor type Ⅰ (FcγR Ⅰ ) protein expression in dorsal root ganglion (DRG) in rats and discuss the mechanism of FcγR Ⅰ in DRG on neuropathic pain preliminary. Methods Thirty-five female Wistar rats were randomly divided into seven groups (n=5): Control group, Sham 3d,7d,14d group and CCI 3d,7d,14d group. Appraisal paw withdrawal mechanical threshold (PWMT) and paw withdrawal thermal latency (PWTL) of the rats before the surgery and 1,3,7,10,14 days later. Then detect FcγR Ⅰ protein expression level in DRG at different time points after the surgery by Western Blotting. Results There was no significant difference in PWMT/PWTL and FcγRⅠprotein expression between the Control and Sham groups(P>0.05). But the PWMT/PWTL was significantly decreased (P<0.01) in CCI group as wel as the up-regulation of FcγR Ⅰ protein expression (P < 0.05) in DRG neurons. Conclusion The up-regulation of FcγR Ⅰ protein expression in DRG might play an important role in neurons excitability increase which was due to the emerging and developing of neuropathic pain.
