Astragalts Polysaccharides%ovariectomy%osteoporosis
目的:评价黄芪多糖预防大鼠卵巢切除后骨质疏松的药效学.方法:取8月龄雌性SD大鼠44只,随机分为4组,每组11只.黄芪多糖组(APS组)大鼠切除双侧卵巢后,黄芪多糖灌胃12周,与单纯卵巢切除组(OVX组)、假手术组(SHAM组)及尼尔雌醇灌胃组(E3组)比较.采用血尿生化检测、骨干/湿重、骨生物力学、骨密度测量和骨形态计量学等方法对黄芪多糖的药效学进行综合评价.结果:与SHAM组比较,OVX组骨干重、骨灰重、骨钙含量和骨密度、抗弯力、骨小梁体积密度及宽度均明显降低,而成骨细胞指数和破骨细胞指数、尿羟脯氨酸则明显增高.黄芪多糖及E3能使上述指标显著改善,接近正常水平.结论:黄芪多糖具有明显抗去势雌鼠骨质疏松作用.

목적:평개황기다당예방대서란소절제후골질소송적약효학.방법:취8월령자성SD대서44지,수궤분위4조,매조11지.황기다당조(APS조)대서절제쌍측란소후,황기다당관위12주,여단순란소절제조(OVX조)、가수술조(SHAM조)급니이자순관위조(E3조)비교.채용혈뇨생화검측、골간/습중、골생물역학、골밀도측량화골형태계량학등방법대황기다당적약효학진행종합평개.결과:여SHAM조비교,OVX조골간중、골회중、골개함량화골밀도、항만력、골소량체적밀도급관도균명현강저,이성골세포지수화파골세포지수、뇨간포안산칙명현증고.황기다당급E3능사상술지표현저개선,접근정상수평.결론:황기다당구유명현항거세자서골질소송작용.
Purpose:To evaluate the anti-osteoporosis effect of astragalts polysaccharides in ovariectomized female rats. Methods:44 female SD rats aged 8 months were divided randomly into 4 groups:the SHAM group (subjected to sham operation);the OVX group (subjected to bilateral ovariectomy);the E3 group (subjected to bilateral ovariectomy and treated after surgery with nilestriol lmg/kg BW by gavage once a week for 12 weeks), and the APS group (subjected to bilateral ovariectomy and treated after surgery with astragalts polysaccharides 1g/kg BW by gavage once a day for 12 weeks). All the animals were sacrified 12 weeks after the operation. The blood and urine samples were collected for determination of biochemical indices. The humeri and tibiae were prepared for determination of net weight,ash weight,calcium and phosphorus content,bone density, as well as bone histopathology and histomorphometry. The left femora were processed for bone anti-curve power. Results:Compared with the SHAM group,the net weight,ash weight,calcium content, density and bending resistance of bones,the trabecular bone volume and thickness were all significantly reduced in the OVX group, while its osteoblast index,osteoclast index and urine hydroxyproline content were obviously increased. In the E3 group and the astragalts polysaccharides group, these indices were close to normal levels. Conclusion:Astragalts polysaccharides can effectively prevent osteoporosis in ovariectomized rats.
