Epithelial rest of Malassez%Apoptosis%Periodontal development
目的:观察牙周组织形成过程中上皮剩余细胞的组织形态、细胞增殖凋亡.方法:TUNEL(TdT-mediated-dUTP nick end labeling,TUNEL)法原位细胞凋亡检测牙周组织发育中上皮剩余细胞凋亡的情况.SP 免疫组化法检测增殖核抗原(PCNA)和凋亡抑制基因Bcl-2、骨桥素(OPN)和骨涎蛋白(BSP)的表达.结果:牙周膜牙骨质侧可见上皮剩余(the epithelial rest of malassez)沿牙骨质表面成簇分布,根分叉和牙颈部较多,细胞数量不等,可见不规则核,簇内细胞相互重叠、排列无规则,TUNEL 结果示上皮剩余中可找到凋亡阳性表达细胞.免疫组化可见上皮剩余细胞阳性表达 OPN,但 PCNA 和 BSP 在此期上皮剩余细胞中为阴性表达.结论:在牙周发育过程中,伴随牙槽骨的改建和牙周膜纤维束走向的调整,上皮剩余经历塑形过程,细胞凋亡在这一过程中发挥重要作用.上皮剩余细胞的 OPN 阳性表达提示其在此过程中可能参与牙骨质的发育.

목적:관찰아주조직형성과정중상피잉여세포적조직형태、세포증식조망.방법:TUNEL(TdT-mediated-dUTP nick end labeling,TUNEL)법원위세포조망검측아주조직발육중상피잉여세포조망적정황.SP 면역조화법검측증식핵항원(PCNA)화조망억제기인Bcl-2、골교소(OPN)화골연단백(BSP)적표체.결과:아주막아골질측가견상피잉여(the epithelial rest of malassez)연아골질표면성족분포,근분차화아경부교다,세포수량불등,가견불규칙핵,족내세포상호중첩、배렬무규칙,TUNEL 결과시상피잉여중가조도조망양성표체세포.면역조화가견상피잉여세포양성표체 OPN,단 PCNA 화 BSP 재차기상피잉여세포중위음성표체.결론:재아주발육과정중,반수아조골적개건화아주막섬유속주향적조정,상피잉여경력소형과정,세포조망재저일과정중발휘중요작용.상피잉여세포적 OPN 양성표체제시기재차과정중가능삼여아골질적발육.
Objective:To observe the morphology,apoptosis,proliferation and distribution of epithelial rests of Malassez(ERM)and expression of bone-related noncollagenous glycoproteins in ERM,so as to evaluate characters and roles of ERM in the developing periodontium.Method:Apoptosis was identified by the terminal deoxy-transferase(TdT)-mediated dUTP-biotin nick end labeling(TUNEL)method.SP immunohistochemical method was used to detect the expression of bcl-2 protein,PCNA(proliferating cell nuclear antigen),OPN(osteopontin)and BSP(bone sialoprotein)in ERM. Result:ERM were observed as clusters of cells,mostly located in the cervical and furcational regions in the periodontal ligament of mouse molars,next to the cementum surface.The number of cells is uncertain,irregularly shaped nuclei could be found in ERM.Some cells of ERM contained dense round/ovoid structures,strongly stained by hematoxylin.Positive cells of TUNEL and OPN could be found in ERM,but the expression of PCNA and BSP in ERM were negative.Conclusion:The epithelial cells of Malassez’s rests undergo turnover or remodeling,along with reconstruction of alveolar bone and periodontal membrane fiber in the developing periodontium,apoptosis and OPN play important roles during the progress.
