目的:利用 MTT 法检测天然化合物对肿瘤细胞的杀伤作用,寻找有抗肿瘤潜力的先导化合物.方法:药物和细胞按所需浓度于96孔板中,恒温培养48 h,加入5μg/ml MTT 溶液20μl/孔,4 h 后吸弃100μl/孔上清液,加入 MTT 裂解液100μl/孔,孵育过夜,用酶标仪检测各孔吸光值,根据 Reed and Menuch 法计算 IC50值.结果:在对人肝癌细胞株 HepG2的毒性试验中,化合物 Qiu17、Qiu33、Qiu22对细胞的抑制率均略低于50%,而阳性对照药物 DDP 则有明显的抑制肿瘤活性.结论:化合物 Qiu17,Qiu33,Qiu22有一定的的抗肿瘤活性作用性.

목적:이용 MTT 법검측천연화합물대종류세포적살상작용,심조유항종류잠력적선도화합물.방법:약물화세포안소수농도우96공판중,항온배양48 h,가입5μg/ml MTT 용액20μl/공,4 h 후흡기100μl/공상청액,가입 MTT 렬해액100μl/공,부육과야,용매표의검측각공흡광치,근거 Reed and Menuch 법계산 IC50치.결과:재대인간암세포주 HepG2적독성시험중,화합물 Qiu17、Qiu33、Qiu22대세포적억제솔균략저우50%,이양성대조약물 DDP 칙유명현적억제종류활성.결론:화합물 Qiu17,Qiu33,Qiu22유일정적적항종류활성작용성.
Objective:By MTT method for detection of natural compounds of tumor cells,looking for the potential antitumor compounds. Method:According to need drugs and cell concentration 96-well plates in cell cultures together,48h,add 5 mu train ml MTT solution 20 g/l/hole,close out after the 4h suction abandon 100 mu l/hole supernatant fluid,join the MTT cracking 100 mu l/hole,incubation overnight,enzyme standard test each hole suction light,according to Menuch method to calculate the Reed and IC50 alue concentration. Result:For HCC in HepG2 lines of toxicity test, Qiu17 compounds,Qiu33,Qiu22 cells were slightly less than 50% of the inhibition rate,and positive DDP drugs significantly inhibit tumor was living. Conclusion:Qiu17 compounds,Qiu33,Qiu22 have anti-tumor activity.
