目的:探索性研究变性高效液相色谱法在检测晚期肺腺癌人类表皮生长因子受体2突变中的应用价值.方法:应用变性高效液相色谱法(denaturing high-performance liquid chromatography,DHPLC)检测了52例晚期肺腺癌患者的血液样本提取的 DNA 中 HER2基因20号外显子的体细胞非移码插入突变,并结合直接测序进行了验证.结果:在对52例晚期肺腺癌患者血清标本提取的 DNA 检测中,通过野生型样本和未知样本的1∶5混合检测,共检出3例 HER2基因20号外显子突变,经过 DNA 测序验证证实均为 YVMA 非移码插入突变.结论:DHPLC 方法可以有效检测出晚期肺腺癌患者血液样本中 HER2基因突变基因片段,具有显著应用价值.

Objective:To exploratory research denaturing high performance liquid chromatography in detection of advanced lung adenocarcinoma human epidermal growth factor receptor 2 mutations in the application.Method:DHPLC was used to detect somatic HER2 Exon 20 somatic non-frameshifting mutations from 52 advanced lung adenocarcinoma patient serum samples.Result:Screened 52 samples with a mixture of 1:5 wild-type and unknown samples and found 3 HER2 Exon 20 YVMA insersion mutations,which was subsequently confirmed by DNA sequencing.Conclusion:DHPLC can efficiently detect mutated HER2 DNA fragments in serum samples from advanced lung adnocarcinoma patients,which is of prominent application value.
