目的探讨不同龄大鼠脑水肿及补体表达的差异.方法使用不同龄SD大鼠建立脑出血模型,分别观察3、7、14、21d时脑组织水含量,神经功能,补体C3、C9的RNA及蛋白表达情况.结果首先对脑水肿的发生发展规律进行分析,发现老龄大鼠与成年大鼠脑水肿发生时程相仿,但水肿程度老龄大鼠轻于成年大鼠;补体C3及C9均有表达,通过免疫组化检查发现这些补体表达主要集中在血肿周围的脑组织,1周左右表达最强,3周基本消退.老龄大鼠的补体表达情况明显弱于成年大鼠;成年大鼠组神经功能恢复较老龄大鼠组更为良好,可能神经元的再生能力在神经功能恢复中起更主要的作用.结论年龄是脑出血后影响预后重要因素,尽管老龄组脑水肿和免疫反应较轻,但神经功能恢复欠佳.

Objective To explore the difference of encephaledema and complement expression between different ages of rats. Methods Cerebral hemorrhage models were established in SD rats of different ages. The liquid content, nerve function, the expression of RNA and protein of complement C3, C9 were observed on 3、7、14、21d. Results The time courses of encephaledema were similar between the aged rats and adults rats, but the severity of encephaledema of the aged rats was less than that of the adult rats;the RNA and protein of complement C3, C9 had expression in both, which were found mainly at the brain tissue around the hemorrhage lesions, and the expression was highest in about a week after the hemorrhage and disappeared 3 weeks later, the expression in ages rats was weaker in the aged rats than in adult rats;the nerve function recovered better in the adult rats than in the old aged rats, which was possibly due to the neuronal regeneration capacity . Conclusions Age is an important factor for the prognosis of cerebral hemorrhage, although in the brain of the aged, the encephaledema and immunity reaction are less severe, the recovery of nerve function is poor.
