目的观察郁金提取物对疼痛小鼠的干预作用及其机制的探讨.方法采用甲醛实验,观察各组小鼠 I、II 相累计舔足时间;行为学观察后取血处理,用放射免疫法测定外周血清 TNF-α的含量并进行比较.结果单侧足底皮下注射甲醛能使小鼠出现明显的两相伤害性行为反应;郁金提取物能抑制甲醛引起的第Ⅱ时相疼痛行为反应,3个剂量组的镇痛作用和模型组相比,差异均具有统计学意义(均 P<0.01);其中郁金提取物300mg/kg 剂量组镇痛作用与阿司匹林组相比,差异具有统计学意义(P<0.05).各组小鼠外周血清中 TNF-α含量与空白组相比,均有明显升高,差异均有统计学意义(均 P<0.01);与造模组相比,郁金提取物各组均能明显降低小鼠血清 TNF-α含量(均 P<0.01);其中郁金提取物300mg/kg 剂量组降低至正常水平,与空白对照组相比,无统计学差异(P >0.05),甚至明显低于阿司匹林组 TNF-α含量(P<0.05).结论郁金提取物具有明显的镇痛作用,其作用部位可能在外周,镇痛机制可能跟降低炎症细胞因子的表达,参与抑制炎症反应有关.

목적관찰욱금제취물대동통소서적간예작용급기궤제적탐토.방법채용갑철실험,관찰각조소서 I、II 상루계첨족시간;행위학관찰후취혈처리,용방사면역법측정외주혈청 TNF-α적함량병진행비교.결과단측족저피하주사갑철능사소서출현명현적량상상해성행위반응;욱금제취물능억제갑철인기적제Ⅱ시상동통행위반응,3개제량조적진통작용화모형조상비,차이균구유통계학의의(균 P<0.01);기중욱금제취물300mg/kg 제량조진통작용여아사필림조상비,차이구유통계학의의(P<0.05).각조소서외주혈청중 TNF-α함량여공백조상비,균유명현승고,차이균유통계학의의(균 P<0.01);여조모조상비,욱금제취물각조균능명현강저소서혈청 TNF-α함량(균 P<0.01);기중욱금제취물300mg/kg 제량조강저지정상수평,여공백대조조상비,무통계학차이(P >0.05),심지명현저우아사필림조 TNF-α함량(P<0.05).결론욱금제취물구유명현적진통작용,기작용부위가능재외주,진통궤제가능근강저염증세포인자적표체,삼여억제염증반응유관.
[ ] Objective To investigate the effects of Radix Curcuma extracts on inflammatory pain in mice and its mecha-nism. Methods The formalin test was adopted to observe the accumulative licking foot times of each phases in mice. The serum levels of tumor necrosis factor-a(TNF-α) were measured by radioimmunoassay. Results Subcutaneous injection of formalin into mouse foot elicited acute two-phase noxious reaction. The second phase of pain reaction induced by formalin was decreased af-ter administration of Radix Curcuma extracts (P<0.01) in a dose-dependent manner. The Radix Curcuma extracts at 300mg/kg showed a better analgesic effect than that of aspirin (P<0.05). The TNF-α level was significant increased in model group com-pared to control group (P<0.01). Radix Curcuma extracts decreased serum TNF-α levels compared to the model group in a dose-dependent manner (P<0.01); TNF-α level in 300mg/kg-dose group was even lower than the baseline level. Conclusion Radix Curcuma extracts have analgesic effects in mice, which may be associated with inhibition of peripheral inflammation.
