目的研究 miR-139-3p 在胃癌组织和癌旁组织中的表达水平及其对 AGS 和 SGC-7901胃癌细胞株的增殖和凋亡的作用.方法采用 qRT-PCR 检测38例患者胃癌组织与癌旁组织中 miR-139-3p 的表达情况,并分析 miR-139-3p 的表达与胃癌临床病理特征之间的关系;利用 miR-139-3p 抑制剂下调胃癌细胞株中 miR-139-3p 的表达,MTT 法检测对 AGS 和SGC-7901细胞增殖的影响,流式细胞仪检测细胞凋亡情况.结果 miR-139-3p 在胃癌组织中表达高于癌旁组织(P<0.01),其表达增高与肿瘤大小和部位相关(均 P<0.05);下调 miR-139-3p 的表达能够抑制胃癌细胞的增殖、促进细胞凋亡(均 P<0.05).结论miR-139-3p 在胃癌组织中表达增高,可能参与胃癌的发生、发展过程.

[ ] Objective To investigate the effect of miR-139-3p on proliferation and apoptosis in gastric cancer. Methods The expression of miR-139-3p in gastric cancer and pericancerous tissues was examined by Real-time PCR. The correlation between miR-139-3p expression and clinicopathologic features of gastric cancer was analyzed. The cultured gastric cancer AGS and SGC-7901 cel s were treated with miR-139-3p Inhibitor; the cel proliferation was detected by MTT assay and apopto-sis was analyzed by flow cytometry. Results The expression of miR-139-3p increased in gastric cancer compared with peri-cancerous tissues (P<0.05). Expression of miR-139-3p was associated with tumor size (P<0.05)and location (P<0.05). Down-regulation of miR-139-3p inhibited proliferation and promoted apoptosis of gastric cancer AGS and SGC-7901 cel s. Conclusion miR-139-3p is up-regulated in gastric cancer, which might be involved in the development of gastric cancer.
