应用比色法、放射免疫测定法(RIA)、连续组织切片及荧光定量RT-PCR技术,检测了不同温度(23℃、26℃、29)℃处理6周对2龄金钱鱼卵巢发育过程中血清中蛋白磷(SPP)、蛋白钙(SPC)、性类固醇激素(雌二醇 E2,睾酮T)及肝和卵巢中卵黄蛋白原mRNA (vtg mRNA)表达的影响.结果显示,实验结束(6周)时,23℃和26℃组卵巢中出现Ⅲ时相卵母细胞,29℃组仅出现Ⅱ时相卵母细胞.除肝体指数(HSI)随处理时间延长而减小外,其他检测指标随时间延长而增加.实验中温度对血清中T水平无显著影响;3周时,23℃和26℃组性腺成熟指数(GSI)及血清中E2水平略高于29℃组,但差异不显著(P>0.05);6周时,两组GSI及血清中E2水平显著大于29℃组(P<0.05);实验期间23℃和26℃组血清中SPP含量显著高于29℃组(P<0.05),而血清SPC含量仅23℃组显著高于29℃组(P<0.05).此外,实验中vtg基因在卵巢的表达量明显低于肝的表达量(P<0.05);肝中vtg表达量与GSI及血清中E2水平变化趋势一致;实验期间卵巢中仅26℃组vtg表达量显著高于29℃组(P<0.05).本研究结果表明,金钱鱼卵巢发育的适宜温度范围为23~26℃,在此温度范围内vtg在肝中的表达量与血清E2含量存在显著的正相关(P<0.05).

We evaluated the effects of temperature on ovarian development in 2-year-old female Scatophagus ar-gus. Fish were reared at 23, 26, and 29℃ for 6 weeks. We measured the levels of serum sex steroid hormones (estradiol-17β, E2;testosterone, T), protein phosphorus (SPP), and protein calcium (SPC). In addition, we quanti-fied vitellogenin (vtg) mRNA expression in the liver and ovary. We observed phase III oocytes in the ovaries of fish reared at 23℃ and 26℃. In contrast, the oocytes in the group reared at 29℃ did not develop beyond phase II by the end of the experiment (6 weeks). The majority of the indices increased with treatment time, except the hepatosomatic index (HSI). Temperature had no effect on the levels of testosterone. The gonadosomatic index (GSI) and E2 levels were significantly elevated in fish held at 23℃ and 26℃ compared with those held at 29℃after 6 weeks (P<0.05). The levels of SPP were higher in fish held at 23°C and 26℃ than in those held at 29℃group, but the difference was only significant for the group held at 23℃ (P<0.05). The level of vtg mRNA ex-pression was lower in the ovary than in the liver. The effect of temperature on liver vtg expression was similar to that on GSI and E2 levels. However, ovarian vtg mRNA expression was only significantly higher in the 26℃group compared with the 29℃ group. In summary, the optimal temperature for the ovarian development of S.argus is between 23 and 26℃. In addition, there is a significant positive correlation between liver vtg mRNA expres-sions and serum E2 content.
