目的 评估突触后致密物(PSD)95多肽疫苗对神经病理性痛大鼠的镇痛效应.方法 用SD大鼠制备保留性神经损伤(SNI)模型,随机分为PSD95组、PBS组和钥孔虫戚血蓝蛋白(KLH)组. PSD95组注射3次PSD95多肽疫苗50μl,后两组分别给予PBS或KLH作为对照.分别于制备SNI前1d、第1次免疫前1d及第3次免疫后14d时测定术侧机械痛阈;并于第1次免疫前1d及第3次免疫后14d时取脑脊液、血清,测定PSD95抗体滴度;测定脊髓背角PSD95蛋白表达水平.结果 PSD95组第1次免疫前1d时未检测到PSD95抗体,而第3次免疫后14d时检测到抗体.与SNI前比较,各组机械痛阈均降低(P<0.05);PSD95组第3次免疫后机械痛阈较其余两组升高,PSD95蛋白表达下调(P<0.05).结论 PSD95多肽疫苗对神经病理性痛大鼠具有镇痛作用,其机制可能与进入脊髓的PSD95抗体下调了PSD95有关.

PSD95組註射3次PSD95多肽疫苗50μl,後兩組分彆給予PBS或KLH作為對照.分彆于製備SNI前1d、第1次免疫前1d及第3次免疫後14d時測定術側機械痛閾;併于第1次免疫前1d及第3次免疫後14d時取腦脊液、血清,測定PSD95抗體滴度;測定脊髓揹角PSD95蛋白錶達水平.結果 PSD95組第1次免疫前1d時未檢測到PSD95抗體,而第3次免疫後14d時檢測到抗體.與SNI前比較,各組機械痛閾均降低(P<0.05);PSD95組第3次免疫後機械痛閾較其餘兩組升高,PSD95蛋白錶達下調(P<0.05).結論 PSD95多肽疫苗對神經病理性痛大鼠具有鎮痛作用,其機製可能與進入脊髓的PSD95抗體下調瞭PSD95有關.
목적 평고돌촉후치밀물(PSD)95다태역묘대신경병이성통대서적진통효응.방법 용SD대서제비보류성신경손상(SNI)모형,수궤분위PSD95조、PBS조화약공충척혈람단백(KLH)조. PSD95조주사3차PSD95다태역묘50μl,후량조분별급여PBS혹KLH작위대조.분별우제비SNI전1d、제1차면역전1d급제3차면역후14d시측정술측궤계통역;병우제1차면역전1d급제3차면역후14d시취뇌척액、혈청,측정PSD95항체적도;측정척수배각PSD95단백표체수평.결과 PSD95조제1차면역전1d시미검측도PSD95항체,이제3차면역후14d시검측도항체.여SNI전비교,각조궤계통역균강저(P<0.05);PSD95조제3차면역후궤계통역교기여량조승고,PSD95단백표체하조(P<0.05).결론 PSD95다태역묘대신경병이성통대서구유진통작용,기궤제가능여진입척수적PSD95항체하조료PSD95유관.
@@@@ Objective To evaluate the analgesic effect of postsynaptic density-95 peptide as subunit vaccine against neuropathic pain in rats.Methods SD rats were randomly divided into group PSD95,group PBS and group KLH afrer spared nerve injury (SNI).The PSD95 group was immunized with subcutaneous PSD 95 peptide vaccine 3 times.Group PBS and KLH served as control groups .Threshold to noxious mechanical stimulation was measured 1day before SNI surgery(baseline),1 day before the 1st immunization and 14 days after the 3rd immunization.Blood and cerebral spinal fluid (CSF) samples were obtained 1day before the 1st immunization and 14 days after the 3rd immu-nization for detection of PSD95 antibody.The expression of PSD95 protein of the lumbar segment of rat spinal cord was determined .Results PSD95 antibody was detected in CSF at 14d after the 3rd immunization in group PSD95.The threshold to mechanical stimulation was signifi-cantly decreased after SNI as compared with the baseline values in all 3 groups.The paw withdrawal threshold to mechanical stimulation after the 3rd immunization was significantly higher and the expression of PSD 95 protein in the dorsal horn of the spinal cord was significantly lower in PSD95 group than in PBS and KLH group.Conclusion PSD95 peptide has analgesic effect on neuropathic pain ,which could be related to the downregulated expression of PSD95.
