目的:观察葛根素对大鼠局灶性脑缺血再灌注后脑组织基质金属蛋白酶-9(MMP-9)表达和脑组织含水率变化的影响,探讨葛根素脑保护的可能机制.方法:54只雄性SD大鼠分为假手术组、对照组和药物组,每组18只.采用线栓法建立右侧大脑中动脉闭塞大鼠模型,时间点为再灌注48h,采用比色法测定脑组织中伊文思蓝(EB)含量反映血脑屏障通透性;干-湿重法检测脑组织含水率;免疫组化法检测脑组织MMP-9表达.结果:脑缺血再灌注后大鼠脑缺血区MMP-9表达明显增加(P<0.05),脑组织含水率明显升高(P<0.05).与对照组比较,药物组大鼠脑组织MMP-9明显低表达(P<0.05),脑组织含水率明显减少(P<0.05).结论:葛根素可能通过抑制MMP-9表达来减轻血脑屏障破坏和脑水肿而发挥缺血再灌注损伤时的脑保护作用.

Objective: To investigate the influence of puerarin on activation of matrix metalloproteinase-9 (MMP-9) in brain tissue and brain edema after cerebral ischemia-reperfusion in rats. Methods: A total 54 rats were randomly assigned to sham-operated(sham) group,control group,and medicine group.Puerarin was given at dose of 70 mg/kg to the medicine group 1 h before ischemia and every 8 h afterwards,and the equivalent amount of normal saline was given to the sham and control groups. The middle cerebral artery of the rats was occluded and recirculated at 48 h. At this time point, the permeability of blood-brain barrier was evaluated based on the leakage of Evans blue. Cerebral edema formation was determined by brain water content. The ex?pression of MMP-9 was measured by immunohistochemistry. Results: Immunohistochemistry revealed an increase of MMP-9 at 48 h post-reperfusion in the control and medicine groups(P<0.05). Puerarin decreased the level of MMP-9 and brain water content in the medicine group as compared to those in the control group(P<0.05). Conclusion: Puerarin may attenuate ischemia-reperfusion injury through inhibition of MMP-9 activation and brain edema.
