[目的]采用OB吻合胶粘贴法建立615小鼠胃癌原位移植瘤模型.[方法]无菌条件下用培养好的小鼠前胃癌细胞株(MFC)制成浓度为1×107个/mL的细胞悬液接种于615小鼠腋下,建立小鼠胃癌皮下移植瘤模型,之后利用皮下移植瘤进行传代,最后采用OB吻合胶粘贴法建立615小鼠胃癌原位移植瘤模型.并设置假手术组,采用形态观察和HE染色方法评价成瘤情况.[结果]形态学观察,模型组10只615小鼠全部成瘤,成瘤率为100%,平均瘤重为1.32±0.64g;假手术组没有肿瘤长出,两组之间有显著性差异(P<0.01).病理结果显示,模型组肿瘤组织细胞体积较大,形状不规则,核质比也较大,是典型的胃癌组织的形态特征.[结论]采用OB吻合胶粘贴法建立615小鼠胃癌原位移植瘤模型建模成功率高、方便、高效、值得进一步研究.

Objective]To establish the model of orthotopic transplanted cancer of gastric tumor in 615 mice with OB anastomotic glue pasted method. [Methods]The subcutaneous transplanted tumor model of gastric cancer was established by injecting cultivated good mice gastric cancer cel line(MFC) into 615 mice with the suspension concentration of 1 ×107/mL sterile tactical y. The subcutaneous transplantation tumor was represented at a time, and the model of orthotopic transplanted cancer of gastric tumor in 615 mice was established with the OB anastomotic glue pasted method. At the same time, sham-operated group was set up. The model was evaluated by morphological observation and HE staining method. [Results] Al 10 tumors of model group were formated, and the tumor formation rate was 100%;the average tumor weight was 1.32 ±0.64g, compared to the sham-operated group without any tumor, there were significant differences; Pathological results showed, the tissue cel s were larger, the shape was irregular,the ratio of nucleus and cytoplasm was larger, these were the typical morphological features of gastric carcinoma. [Conclusion] There wil be the low mortality rate and the high successful rate to establish the model of orthotopic transplanted cancer of gastric tumor in 615 mice with OB anastomotic glue pasted method, and it wil be simple and efficient and worth further research.
