采用微波提取的方法,从中药材金线莲中提取总生物碱,以盐酸麻黄碱为对照品,分光光度法测定了总生物碱含量。以总生物碱含量为考察指标,讨论提取料液比、提取功率、提取时间等因素对提取结果的影响。通过正交试验L9(33),对提取条件进行优化,得到微波提取的最佳工艺条件,即料液比为1∶15 g/mL,提取功率为616 W,提取时间为6 min。与传统的加热回流法对照,经F-检验法及t-检验法检验,测定结果没有显著性差异。实验表明,该法操作简便,大大缩短了提取时间,节约了人力、物力,提取效果令人满意,对金线莲生物碱的开发利用提供了借鉴。
