目的:观察益母草总黄酮对去卵巢大鼠下丘脑和海马雌激素受体α及β mRNA表达的影响,探讨其对绝经后骨质疏松的作用机制。方法选取10~11月龄SD雌性大鼠48只,随机分为对照组、去势组、益母草总黄酮组和雌二醇组,以双侧卵巢切除法建立大鼠骨质疏松模型。益母草总黄酮组、雌二醇组分别用益母草总黄酮、雌二醇给药4个月,采用RT-PCR法检测各组下丘脑和海马雌激素受体α及β mRNA的表达。结果大鼠卵巢切除后,血清中雌二醇水平、椎骨骨密度、子宫湿重、下丘脑和海马雌激素受体α及βmRNA的表达均明显下降。雌二醇治疗后血清中雌二醇水平、椎骨骨密度、子宫湿重升高,下丘脑和海马β mRNA的表达均明显下降。益母草总黄酮治疗后,血清中雌二醇水平、椎骨骨密度、下丘脑和海马雌激素受体α及β mRNA的表达均明显升高,子宫湿重无明显改变。结论益母草总黄酮对切除卵巢所致的骨质疏松大鼠有防治作用,对子宫无不良影响。

목적:관찰익모초총황동대거란소대서하구뇌화해마자격소수체α급β mRNA표체적영향,탐토기대절경후골질소송적작용궤제。방법선취10~11월령SD자성대서48지,수궤분위대조조、거세조、익모초총황동조화자이순조,이쌍측란소절제법건립대서골질소송모형。익모초총황동조、자이순조분별용익모초총황동、자이순급약4개월,채용RT-PCR법검측각조하구뇌화해마자격소수체α급β mRNA적표체。결과대서란소절제후,혈청중자이순수평、추골골밀도、자궁습중、하구뇌화해마자격소수체α급βmRNA적표체균명현하강。자이순치료후혈청중자이순수평、추골골밀도、자궁습중승고,하구뇌화해마β mRNA적표체균명현하강。익모초총황동치료후,혈청중자이순수평、추골골밀도、하구뇌화해마자격소수체α급β mRNA적표체균명현승고,자궁습중무명현개변。결론익모초총황동대절제란소소치적골질소송대서유방치작용,대자궁무불량영향。
Objective To explore the effect of total flavonoids of Herba leonuri( TFH) on the mRNA expressions of estrogen receptor alpha ( ERα) and estrogen receptor beta ( ERβ) in hippocampus and hypothalamus in ovariectomized ( OVX ) Sprague-Dawley ( SD ) rats, and the mechanism of TFH against postmenopausal osteoporosis. Method Forty-eight female SD rats(10~11 months) were randomly divided into 4 groups:a sham group,an ovariectomy group,a TFH group,and an estradiol group. The RT-PCR was performed to determine the mRNA expression of ERα and ERβ in hypothalamus and hippocampus. Results After the ovariectomy, the serum estradiol level,bone mineral density( BMD) of vertebra,wet weight of uterus,and the mRNA expressions of ERα and ERβ in hypothalamus and hippocampus were markedly decreased in OVX rats;after the treatment with TFH,the serum estradiol level,bone mineral density(BMD) of vertebra,wet weight of uterus,and the mRNA expressions of ERα and ERβ in hypothalamus and hippocampus were increased except that the wet weight of uterus did not change. Conclusion TFH can improve BMD of vertebra in the OVX rats without untoward effects on the uterus.
